“…In normal rats, pharmacokinetic profile of pure baicalin and baicalin-contained KOB extracts showed two plasma concentration peaks of baicalin (Figure 3). It was reported that generally flavone glucuronides show bimodal pharmacokinetic profile (Lu et al, 2007;Tong et al, 2012). The first absorption occurred immediately within 1 h. This absorption was considered owing to the direct absorption.…”

“…The second absorption occurred at 8-12 h. This absorption probably occurred through glucuronidation. During glucuronidation, enteric circulation and enterohepatic circulation may affect the second peak (Lu et al, 2007;Tong et al, 2012). When standard baicalin was administered orally, normal rats showed 0.683 mg/ml of C max and normal rats administered with KOB extracts showed 0.457 mg/ml of C max .…”

“…Hence, such standard samples lack compatibility with the test samples and they may lead to deceptive results and make the method validity questionable. Unfortunately, this was widely done in methods published earlier (10,(16)(17)(18)(19). Such approaches are in contradiction with the recommendations of the FDA guidance for bioanalytical method validation, which state that the concentrations of standards should be chosen on the basis of the concentration range expected in a particular study (12).…”

“…This can further explain the high recovery of all concentrations of wogonin from group 1 and potentiates our hypothesis of such samples being incompatible with the test samples. Analysis of previous methods of wogonin quantitation in plasma showed that changing the plasma matrix with organic solvents was widely conducted during standard sample preparations (7,8,10,(16)(17)(18)(19)21). It was performed through direct spiking of the working standard stock solutions of wogonin (with organic solvents) in ratios ranging between 10-50 % of the blank plasma volume while ignoring the consequences for drug solubility and sample integrity and hence their validity.…”

“…Recently, the biological activities of Scutellariae Radix have been reported on anti-bacterial effect (Yun et al, 2012), anti-virus (Moghaddam et al, 2014), anti-inflammation (Kim et al, 2009), anti-allergy (Hsieh et al, 2007, sedation (Park et al, 2006), anti-hypertensive effect (Song et al, 2013), and antioxidation (Cao et al, 2011). In the chemicophysical study of secondary metabolites in natural products, baicalein (Horvath et al, 2005), baicalin (Ohkoshi et al, 2009), chrysin (Tong et al, 2012), oroxylin-A (Ku et al, 2014), oroxylin-α-7-O- show differently effects on anti-oxidation and neuro-protective activity . However, we couldn't find the report on effective extraction method of enriched flavonoid fraction for industrialization of Scutellariae Radix.…”
