Simplified Sizing of Low-Density Lipoprotein Using Polyacrylamide Gradient Gel Electrophoresis of Plasma

Abstract: Low-density lipoprotein (LDL) particles can be separated into subtractions according to size by nondenaturing polyacrylamide gradient gel electrophoresis. Established research methods require specialised equipment and are frequently unsuited to the clinical laboratory. In this study, we utilised a colour flat bed scanner in conjunction with shareware image analysis software to compare LDL particle diameters of isolated LDL with LDL in whole plasma. LDL was isolated by ultracentrifugation and electrophoresed on… Show more

“…Although Photoshop has a range of commands for adjusting the tonal quality and color separation of images, only a few reports in the literature have applied it to biomedical research. Several authors have used Photoshop only as a way of scanning images into their computers for later analysis by other programs 29–32 . Others have used Photoshop to superimpose grids over scanned images 33 or to assess facial movement as a means of grading facial nerve injury 34 .…”

“…There are only a few reports in the literature in which Photoshop has been applied to biomedical research. Several authors have used Photoshop merely as a way of importing scanned images into their computers for later analysis by other programs (Brown et al 1995; Westhuyzen et al 1997). For example, Ikeda and coworkers (1997) have used Photoshop not only to scan selected fields from microscope slides but also to compose seamless widefield images of large tissue sections.…”

“…During routine quality control tests, the intra-assay coefficient of variation for Rf ranged from 1.6 % to 3%. There is no criterion measure of LDL particle size, but polyacrylamide gel electrophoresis and ultracentrifugation have shown high agreement [6,36]. Polyacrylamide gel electrophoresis has also been used effectively in demonstrating that LDL particle size is inversely related to CHD risk and thus can be said to have face validity [4,18].…”
