Simplified scheme for routine identification of human Staphylococcus species

Abstract: From a total of 40 characters that were previously used to differentiate species of staphylococci, 13 key characters were selected to make a simplified scheme that could be easily used by the routine clinical laboratory for identifying human staphylococci. These key characters included coagulase activity, hemolysis, nitrate reduction, and aerobic acid production from fructose, xylose, arabinose, ribose, maltose, lactose, sucrose, trehalose, mannitol, and xylitol. In the simplified scheme, 924 strains of staphy… Show more

“…intermedius were differentiated from Staph. aureus by lack of pigment production (Kloos and Schleifer 1975), lack of acid production from D-manniotol (Sneath 1986), lack of betahaemolysis and variability with the slide test (Balows 1991).…”

“…It is usual to use the classification of Kloos et al (1974) and Kloos & Schleifer (1975), which lists 10 species of Staphylococcus and seven species of Micrococcus. The International Subcommittee for Staphylococcus Taxonomy (Hill 1976) recommends a simplified scheme based on the work of these authors.…”

“…The International Subcommittee for Staphylococcus Taxonomy (Hill 1976) recommends a simplified scheme based on the work of these authors. A modified Kloos & Schleifer (1975) scheme for staphylococci was used successfully by Eady (1979; A scheme of lyogroups based on the ability of staphylococci to lyse heat-killed Mic. luteus ATCC 4698 in various media agrees well with the above taxonomy and claims to be better adapted for routine use Varaldo et al , 1980.…”

“…All samples were cultivated within an hour as follows: The urinary samples (10 ml) were centrifuged (1000 g, 10 min at 4OC) and the pellets were cultivated on Columbia agar (BBL, Cockeysville, Md., USA) containing 5 O/O lactose (Stock Solution 20 O,' O wiv) and phenol red and Identificaticn of the aerobic and anaerobic bacteria to the genus level was made according to Cowan & Steel (1974). Coagulase-negative staphylococci were typed to species level according to the scheme of Kloos & Schleifer (1975). Staphylococcus aureus were phage-typed with a standard set G f phages (Blair & Williams 1961).…”

“…When 0.01 ml samples of suspensions of coagulase-negative staphylococci (106 bacteriaiml) or lactobacilli (106 bacteria/ml) were incubated in 1 ml fresh semen for 18 h in 3 7 O C the reccvery was a tenfold increase. 2) Coagulase-negative staphylococci found from patients 1 and 2 were typed to species level according to Kloos and Schleifer (1975).…”
