Summary Plants are simultaneously attacked by different pests that rely on sugars uptake from plants. An understanding of the role of plant sugar allocation in these multipartite interactions is limited. Here, we characterized the expression patterns of sucrose transporter genes and evaluated the impact of targeted transporter gene mutants and brown planthopper (BPH) phloem‐feeding and oviposition on root sugar allocation and BPH‐reduced rice susceptibility to Meloidogyne graminicola. We found that the sugar transporter genes OsSUT1 and OsSUT2 are induced at BPH oviposition sites. OsSUT2 mutants showed a higher resistance to gravid BPH than to nymph BPH, and this was correlated with callose deposition, as reflected in a different effect on M. graminicola infection. BPH phloem‐feeding caused inhibition of callose deposition that was counteracted by BPH oviposition. Meanwhile, this pivotal role of sugar allocation in BPH‐reduced rice susceptibility to M. graminicola was validated on rice cultivar RHT harbouring BPH resistance genes Bph3 and Bph17. In conclusion, we demonstrated that rice susceptibility to M. graminicola is regulated by BPH phloem‐feeding and oviposition on rice through differences in plant sugar allocation.

Background: Laodelphax striatellus transmits rice stripe virus (RSV) during sap feeding on the rice plant. The insect saliva proteins have direct and indirect roles in mediating RSV transmission; however, the function of most saliva proteins remains unclear.Results: In this study, we sequenced L. striatellus saliva proteins using shotgun liquid chromatography-electrospray ionizationtandem mass spectrometry. We identified 41 secreted saliva proteins, among which a saliva mucin-like protein, designated LssaMP, was the most abundant. In silico analysis revealed the sequence conservation among planthoppers. We revealed that the LssaMP gene is specifically expressed in the salivary glands and the protein is secreted as a component of gel saliva. Using LssaMP-specific double-stranded RNA (dsRNA) to silence gene expression, we revealed that LssaMP is required for formation of the salivary sheath, an important structure for sap feeding. Disrupting LssaMP expression resulted in inefficient formation of the feeding structure, thereby stopping insects from secreting watery saliva and acquiring sufficient nutrients from the phloem sap. We confirmed that RSV is mainly released via the watery saliva, which passes through the salivary sheathes into the plant phloem. An insufficient feeding structure results in decreased release of watery saliva, as well as the arboviruses.Conclusion: This study clarified the function of an insect saliva protein in mediating insect feeding, as well as arbovirus transmission.

Trichomes on Cotton Leaf Surface Affect the Feeding Behaviors of Cotton Aphids

Aphis gossypii Glover is one of the most important pests in cotton seedling. The specialized mouthpart and short generation time allow them seriously restricted the quality and yield of cotton. With the electrical penetration graph (EPG), optical microscope, and scanning electron microscope, the physical characteristics of trichomes on the cotton leaf surface and their effects on the feeding behaviors of cotton aphids were investigated in this study. Two types of leaf trichomes, glandular trichomes, and asteroid non-glandular trichomes were observed on the surface of cotton leaves under optical and scanning electron microscopes. As a whole, EPG results mainly showed differences in a non-probe waveform which meant searching on the leaf surface, and a potential drop waveform which meant intracellular penetration. Whereas pathway waveform, xylem waveform, and phloem waveform showed inconspicuous differences. Detailed indicators were slightly different at different periods. Our results confirmed that the structure of trichomes may directly affect the searching behaviors of herbivorous insect pests and hence regulate the plant-environment interactions.
