Simple and on‐site DNA purification for LAMP reaction applicable to non‐adult tephritid fruit fly (Diptera: Tephiritidae)

Abstract: Tephritid fruit flies are important pests of fruits and vegetables in Southeast Asia and the Pacific region (Drew & Romig, 1997). Among them, the Bactrocera dorsalis complex comprises the oriental fruit fly B. dorsalis Hendel (Diptera: Tephiritidae) and its sibling species, which are economically important and extremely polyphagous (Clarke et al., 2005; Drew & Hancock, 1994). Many studies have examined control of B. dorsalis complex in invaded areas (e.g. Armstrong

“…The quick DNA extraction method was first applied to Plasmodium falciparum in LAMP assay using a heat‐based (99℃ for 10 min) treatment to prepare DNA for LAMP rapidly, and compared with the column or organic solvent‐based DNA extraction method (Poon et al., 2006). In 2020, quick and simple DNA preparation was applied to fruit flies for the first time with the improved ‘toothpick method’ adapted from Fukuta et al (2005) (Kitano & Takakura, 2020). The ‘toothpick method’ was applied and improved in our research.…”

“…To prepare DNA of target species quickly, sterile toothpicks were used for quick extraction method modified from Kitano and Takakura's method (2020). First, living larvae and pupae were washed with sterilized distilled water, and the insects were removed onto a filter paper to dry.…”

“…These were commercialized by Optigene ( http://www.optigene.co.uk/plant-health-pests-diagnostics ) and are used for species identification at two ports of entry in Switzerland ( von Felten, 2017 ). For a similar purpose, a LAMP kit was developed for rapid identification of Bactrocera flies ( Bactrocera dorsalis complex, Bactrocera cucurbitae / Bactrocera latifrons and Bactrocera correcta / Bactrocera zonata ) ( Blaser et al., 2018 ), while recently a LAMP identification method was reported for B. tryoni ( Blacket et al., 2020 ), Dacus ciliatus ( Sabahi et al., 2018 ) and Zeugodacus scutellatus ( Kitano & Takakura, 2020 ). In 2009, Huang et al.…”
