Silica nanoparticles induce neurodegeneration-like changes in behavior, neuropathology, and affect synapse through MAPK activation

Abstract: BackgroundSilica nanoparticles (SiO2-NPs) are naturally enriched and broadly utilized in the manufacturing industry. While previous studies have demonstrated toxicity in neuronal cell lines after SiO2-NPs exposure, the role of SiO2-NPs in neurodegeneration is largely unknown. Here, we evaluated the effects of SiO2-NPs-exposure on behavior, neuropathology, and synapse in young adult mice and primary cortical neuron cultures.ResultsMale C57BL/6 N mice (3 months old) were exposed to either vehicle (sterile PBS) o… Show more

“…The data from the WT mice were displayed on the left side of the dotted line, and the Trpm2 −/− data on the right side. * P < 0.05 compared with the control group thought close to that exposed to human population [53]. As has been well documented, immune response is a complex process, composed of recruitment of many types of immune cells and subsequent generation of pro-inflammatory cytokines and chemokines at different time courses.…”

“…Many studies included six animals/group for toxicological experiments. [11][12][13][14] The concentration of each pollutant was fixed according to the international safety standards specified by WHO. This subacute toxicity model lasted for 30 days and received intra-tracheal instillation for 10 times before being executed.…”

“…The applied dose of SiNPs was in reference to an inhalation study in mice [84], in which the doses of SiNPs were evaluated based on the real workplace exposure scenarios. It was reported the occupational exposure level of SiNPs ranged from 1.0-27.6 mg/m 3 [85].…”
