Silica-based nanoparticles for photodynamic therapy applications

Abstract: Silica-based nanoparticles for applications in photodynamic therapy (PDT) have emerged as a promising field for the treatment of cancer. In this review, based on the pathway the photosensitizer is entrapped inside the silica matrix, the different methods for the synthesis of silica-based nanoparticles are described from the pioneering works to the latest achievements which concern multifunctional nanoplatforms, up-converting nanoparticles, two-photon PDT, vectorization and in vivo applications.

“…It was found that both PpIX-Ole-DOPC and PpIX-Ole are much more effective than the non-functionalized PpIX and the results were due to the drug delivery characteristic of the liposome, which showed an effective role in endocytosis. In the same study it was also reported (Couleaud et al 2010, Lucky et al 2015, Piao et al 2008.…”

“…Moreover, ORMOSIL can possess functional groups added to the surface that make these SiNPs promising vectors for PDT applications (Couleaud et al 2010). For instance, Ohulchanskyy et al were able to prepare ORMOSIL NPs with a PS molecule covalently incorporated into the SiNP aiming to minimize the PS release during systemic circulation (Ohulchanskyy et al 2007).…”

“…Such drawbacks of molecular photosensitisers for PDT can be overcome through the use of nanoparticles as drug delivery systems. 5 Silica nanoparticles, 6 polymeric nanoparticles, 7 magnetic nanoparticles, 5,7 quantum dots, 8 upconverting nanoparticles 9 and especially gold nanoparticles 5 are emerging as ideal candidates for use as delivery vehicles of hydrophobic photosensitisers for PDT. In the field of gold nanoparticles (AuNPs), Brust and co-workers reported a method based on the use of tetraoctylammonium bromide as the phase transfer reagent for the synthesis of gold nanoparticles functionalised with hydrophobic ligands that were ultimately soluble in polar solvents.…”

“…Photodynamic therapy (PDT) is another important emerging research field for the development of nanoscale therapeutics (Bechet et al, 2008;Couleaud et al, 2010b;Vanderesse et al, 2011) that can optimize drug delivery of materials or systems. Photodynamic therapy involves the use of light, photosensitizers and oxygen.…”
