Silencing a cotton bollworm P450 monooxygenase gene by plant-mediated RNAi impairs larval tolerance of gossypol

Abstract: We identify a cytochrome P450 gene (CYP6AE14) from cotton bollworm (Helicoverpa armigera), which permits this herbivore to tolerate otherwise inhibitory concentrations of the cotton metabolite, gossypol. CYP6AE14 is highly expressed in the midgut and its expression correlates with larval growth when gossypol is included in the diet. When larvae are fed plant material expressing double-stranded RNA (dsRNA) specific to CYP6AE14, levels of this transcript in the midgut decrease and larval growth is retarded. Both… Show more

“…Higher mortality was observed in larvae reared on a plant line expressing higher levels of unprocessed, full-length HaAK dsRNA . Arabidopsis thaliana triple mutants lacking dcl2, dcl3 and dcl4 produced more intact, long hairpin RNA and caused more pronounced silencing of CYP6AE14 compared to CYP6AE14-expressing control plants with lower levels of intact dsRNA (Mao et al, 2007). Efficacy of longer, undiced dsRNA on…”

“…housekeeping genes), genes with high mRNA turnover coding for proteins with short half-life, stability of dsRNA, and the ability to deliver sufficient amounts of dsRNA (Huvenne and Smagghe, 2010;Scott et al, 2013). The potential applications for RNAi in pest management include oral administration of dsRNA in baits (Zhou et al, 2008), embedded in nanoparticles (Zhang et al, 2010), expressed in E. coli and formulated as a biological insecticide (Zhu et al, 2011), or expressed in transgenic plants (Baum et al, 2007;Mao et al, 2007).…”
