Significant Amelioration of Tracheal Stenosis following Lenvatinib in a Patient Who Has Anaplastic Thyroid Carcinoma with Bronchomediastinal Infiltration: A Case Report

Abstract: Background: Anaplastic thyroid carcinoma has an extremely poor prognosis, and no known drugs have exhibited acceptable efficacy. In recent years, novel anticancer tyrosine kinase inhibitors have been developed. We encountered a case of tracheal stenosis due to mediastinal and tracheal infiltration of anaplastic carcinoma for which lenvatinib exhibited remarkable effects; owing to this, airway management could be performed, even though the patient’s condition was considered critical. Case Report: A 55-year-old … Show more

“…Also a case report from Japan showed significant amelioration of tracheal stenosis following lenvatinib in a patient who had ATC with bronchial and mediastinal infiltration. The patient had failed radiation and chemotherapy prior to receiving lenvatinib (30). A phase II trial of lenvatinib, an open label, single arm, multi-center study for the treatment of ATC was prematurely stopped due to not reaching the minimum outcome expected.…”

“…In the present case, we repeatedly administered lenvatinib to the patient despite severe side effects, including PRES, because we thought that lenvatinib would prolong her survival. It was recently reported that lenvatinib treatment was effective for locally advanced ATC and led to an improved quality of life as well as prolonged survival in Japanese patients ( 2 , 23 ). As previously mentioned, most ATC patients die within 6 months of the diagnosis ( 1 - 3 ).…”

“…Recent anecdotal evidence and a phase II clinical study have reported the antineoplastic activity of lenvatinib in ATC (16)(17)(18)(19)(20). In the present study, we aimed to evaluate the antineoplastic activity of lenvatinib in ATC continuous cell lines and in primary ATC cell cultures both in vitro and in vivo.…”
