Signaling, metabolism, and cancer: An important relationship for therapeutic intervention

Abstract: In cancerous cells, significant changes occur in the activity of signaling pathways affecting a wide range of cellular activities ranging from growth and proliferation to apoptosis, invasiveness, and metastasis. Extensive changes also happen with respect to the metabolism of a cancerous cell encompassing a wide range of functions that include: nutrient acquisition, biosynthesis of macromolecules, and energy generation. These changes are important and some therapeutic approaches for treating cancers have focuse… Show more

“…Various studies have shown that some signaling pathways can be targeted using CRISPR/Cas9 technology and can lead to attenuation of drug resistance in cancer cells. One of the most important of these pathways is the PI3K/AKT signaling pathway, which is involved in enhancing proliferation, attenuating apoptosis, and enhancing drug resistance [129,130]. In one study, genes encoding PI3K-110α and PI3K-110β (Pik3ca and Pik3cb) were targeted and knocked out using the CRISPR/Cas9 system.…”

“…Active PI3K activates a secondary messenger called PIP3 with phosphorylation of membrane lipids, and this is PIP3 that activates AKT. AKT enhances cell survival, inhibits apoptosis and also increases cell proliferation [ 116 , 117 ].…”

“…Furthermore, the amount of G6PD and TKT enzymes is elevated in colorectal cancer and hepatocellular carcinoma patient tissues compared to normal counterparts [40,42]. Thus, exploiting the reprogrammed metabolism in tumor cells may offer novel therapeutic targets against cancer [9,43]. We will next cover the use of PPP inhibitors as a therapeutic window in cancer cells.…”
