Sialyl Lewisa Expression as a Predictor of the Prognosis of Colon Carcinoma Patients in a Prospective Randomized Clinical Trial

Abstract: This prospective study in a randomized controlled trial suggests that sialyl Lewis(a) expression levels may serve as an indicator of the metastatic potential of colon cancer cells, which would strongly predict the prognosis.

“…Expression of sialyl Lewis a , which is one of the most important molecules of cell surface carbohydrates, was assessed in its putative function as a prognostic marker of aggressive and metastasizing tumor growth. In one of the few randomized studies investigating specimen of 233 patients, this hypothesis was confirmed [31]. The authors concluded that the expression of the sialyl Lewis a antigen may serve as an indicator of the metastatic potential of colon cancer cells, which would strongly predict the prognosis.…”

“…Sialyl-Lewis a (sLe a ; CA 19-9), a mucin epitope whose levels in CRC correlate with overall survival and disease free survival, may serve as an indicator of metastatic potential [141,151]. It is believed that tumour metastasis and cancer cell arrest in the vasculature is facilitated through the tethering of circulating cancer cells to vascular endothelium through the interaction of molecules such as CD15s, CA 19-9 and CD44 variants on the tumour cell surface and selectins (CD62-P, -L and –E) expressed on endothelial cells, neutrophils, monocytes, NK cells or platelets [152155].…”

“…Both, sLe a and sLe x are typical tumor associated carbohydrate antigens and have been implicated with tumor progression and enhanced metastasis formation [40] of different neoplasias including colon and colorectal cancer [41][42][43][44][45][46][47], lung cancer [48][49][50][51], gastric cancer [52,53], pancreatic cancer [54,55], prostate cancer [56,57], and breast cancer [58]. Sialyl Lewis a is a prognostic factor in colon and colorectal cancer whereby increased sLe a levels are associated with a poor prognosis of the afflicted patients [41,42,45].…”
