Shunting Inhibition Improves Robustness of Gamma Oscillations in Hippocampal Interneuron Networks by Homogenizing Firing Rates

Abstract: Networks of GABAergic neurons are key elements in the generation of gamma oscillations in the brain. Computational studies suggested that the emergence of coherent oscillations requires hyperpolarizing inhibition. Here, we show that GABA(A) receptor-mediated inhibition in mature interneurons of the hippocampal dentate gyrus is shunting rather than hyperpolarizing. Unexpectedly, when shunting inhibition is incorporated into a structured interneuron network model with fast and strong synapses, coherent oscillati… Show more

“…Our results show that two distinct types of GABAergic interneuron, PV-expressing basket and ivy cells, the two most numerous GABAergic cell types in the hippocampal pyramidal layer, contribute differentially to behavioral state-dependent neuronal activities and 2,13,15,35,36 through their highly precise inhibitory control of pyramidal cell output 5, 37-39 . They have been predicted to provide a clock-like temporal frame for information coding by pyramidal cells 40 .…”

“…So far, an unequivocal recognition of PVexpressing basket cells is only possible by verifying both their molecular expression and axonal arborizations, because PV is expressed also by hippocampal axo-axonic, bistratifed, O-LM and some long-range projection neurons and because other types of basket cell exist which do not express parvalbumin 34 . Parvalbumin-expressing basket cells are thought to contribute to the generation of network oscillations 2,13,15,35,36 through their highly precise inhibitory control of pyramidal cell output 5, 37-39 . “…It is a pentameric receptor that selectively conducts anions, particularly chloride ions. The activation of GABA A receptors generally causes hyperpolarization of the neuron and shunting inhibition, [115][116][117] which diminishes the chance of a successful firing of action potentials. 118,119 Different anesthetics preferentially target different neurotransmitter receptors.…”

“…For example, a low value of R m , which may be generated by a high density of leak channels (23), would accelerate the decay of EPSPs by speeding up the membrane time constant. Similarly, a low R i value, which may be caused by a high intracellular Cl -concentration in BCs (24), could accelerate dendrosomatic propagation of synaptic signals.…”
