Show Yourself, Asparaginase: An Enzymatic Reaction Explained through a Hands-On Interactive Activity

Abstract: Determining the catalytic activity of an enzyme can be the perfect method for its identification, for example during purification procedures or for isolation purposes. Herein, we used a pharmaceutically relevant protein to bring the concept of enzymatic activity to the classroom. We designed a hands-on interactive activity in which a medically relevant enzyme, asparaginase, was distinguished from a nonenzymatic protein based on its specific enzymatic activity. The experiment was carried out in the classroom, d… Show more

“…Using a pharmaceutically relevant protein, the authors of the other research 55 brought the concept of enzymatic activity to the classroom. They designed a hands-on interactive activity to promote the emerging field of protein-based drugs as a source of scientific careers in nanobiotechnology and to show the students an image of "scientist" as an ordinary person working in an exciting profession.…”

“…If the enzyme is attached to a nonporous material, mass transfer limitations occur only because of the stagnant film, or Nernst layer surrounding the support material. Should the enzyme be entrapped inside a porous matrix, internal mass transfer limitations come into play as well. , Numerous experiments on enzyme catalysis have already been published in this Journal , focusing on fundamental and applied aspects of biocatalysis. While immobilization has been tackled in diverse reports, to the best of our knowledge no previous pedagogical experiment exists pertaining to the evaluation of immobilization on (observed) enzyme kinetics and substrate mass transfer.…”

“…Although bioconjugation reactions have already been presented in a small number of experimental reports in this Journal , none of the previous papers mention the application of protein PEGylation in a lab class. As this technique has industrial importance (mainly for the biopharmaceutical sector), the knowledge and application of this reaction in lab classes contributes to a better understanding of biological drugs with chemical modifications by students.…”
