to the oriental fruit fly, Bactrocera dorsalis (Hendel) (Wan et al. 2011), which limits the development of agriculture in many countries due to reduction in farm income; it also leads to overuse of pesticides. The difficult problems in B. tau control are to rapidly and accurately identify the morphologically similar cryptic species complexes, and reconstruct historical processes to tackle the issue of the origin and range expansion (Dujardin and Kitthawee 2013). Microsatellites, also known as simple sequence repeats (SSRs), are highly polymorphic and codominant molecular markers based on simple repeated and frequent sequences common in eukaryotic genomes, which have proven to be a powerful tool available in genetic diversity, genome mapping, molecular ecology and evolutionary studies (Goldstein and Schlötterer 1999). Microsatellites could solve many of these problems because of the power and ability for investigating population genetic differentiation. Therefore, this study was aimed at developing and characterizing a microsatellite marker set for B. tau. Materials and methods Insect collectionAdult flies were collected from a fruit garden in the suburb of Hainan, China. Flies were captured using traps baited with parakairomone methyl eugenol. B. tau adults were identified to species based on morphology. Construction and sequencing of microsatellite-enriched genomic libraryTotal genomic DNA of B. tau was extracted from the adult individual with standard phenol-chloroform extraction Abstract The fruit fly, Bactrocera tau (Walker), is one of the most important invasive worldwide pests on fruits and vegetables. In order to facilitate species identification and population genetic analysis, we isolated 22 microsatellite loci through an enriched genomic library protocol. Polymorphisms were evaluated in 30 individuals. The number of alleles per locus ranged from 2 to 9, and the observed and expected heterozygosities ranged from 0.048 to 0.862 and 0.136 to 0.852, respectively. The polymorphic information content values were from 0.124 to 0.818. Eight loci had significant deviations from the Hardy-Weinberg equilibrium. These SSR markers can be used to understand the population genetics and evolutionary patterns of the B. tau complex.

The oriental armyworm, Mythimna separata, is a serious agricultural pest in China. Seasonal and roundtrip migration has recently led to sudden, localized outbreaks and crop losses. To evaluate genetic differentiation between populations in eastern and western China and elucidate gene flow, the genetic structure of 20 natural populations from nine provinces was examined using seven microsatellite markers. The results indicated high genetic diversity. However, little to moderate (0 < F ST < 0.15) genetic differentiation was detected, and there was no correlation between genetic distance and geographical distance. Bayesian clustering analysis identified three groups whereas discriminant analysis of principal components identified ten clusters that were considered as two clear‐cut clusters and one admixed group. Gene flow occurred frequently in most population pairs, and an asymmetrical migration rate was detected in several pairwise population comparisons. The bottleneck test showed that few populations had experienced recent bottlenecks. Correspondingly, large‐scale and long‐distance migration of M. separata has caused low genetic differentiation and frequent gene exchange. Our findings are important for studying genetic evolution and help to improve predictions of M. separata outbreaks in China.

With the advent of next‐generation Roche 454 pyrosequencing technologies, transcriptome level sequence collections are arising as prominent resources for the discovery of gene‐based molecular markers. In a previous study more than 1450 simple sequence repeats (SSRs) in expressed sequence tag (EST) sequences resulting from 454 pyrosequencing of Laodelphax striatellus cDNA were identified. From these we developed PCR primers for 40 di‐ or tri‐candidate SSRs (minimum repeats > 8) from the combined EST library. After extensive optimization, 13 pairs were end labeled with a fluorescent dye. Here, we also tested these 13 SSRs for cross‐species amplification in two other planthoppers, Nilaparvata lugens and Sogatella furcifera. The marker transferability was considerably high in N. lugens (92.31%) and in S. furcifera (61.54%). All of the 13 fluorescent SSRs were polymorphic, with allele numbers ranging from two to seven for 24 male and 24 female L. striatellus individuals. Observed heterozygosities ranged from 0.016 to 0.621. Development of SSR markers from ESTs will be a valuable tool to improve our understanding of population structure in this important rice pest.
