Understanding distribution and transport of carbon assimilates and photosynthesis contribution to grain yield in wheat spike is important in assessing the photosynthetic process under stress conditions. In this study, photosynthetic characteristics were evaluated in a pot experiment. Transport of spike photosynthates to grain was demonstrated using 14 C isotope tracer technique. Yield and key enzyme activities of C 3 and C 4 pathways were examined after anthesis in wheat cultivars of different drought resistance. The ear net photosynthetic rate, chlorophyll content of the spike bracts (glume, lemma, and palea), and relative water content slightly decreased under water deficit in drought resistant variety Pubing 143 (Pub) during the grain filling stage, whereas all parameters decreased significantly in drought sensitive variety Zhengyin 1 (Zhe). Grain 14 C-photosynthate distribution rate fell by 3.8% in Pub and increased by 3.9% in Zhe. After harvest, the water-use efficiency of Zhe dropped by 18.7% under water deficit. Rubisco activity in ear organs declined significantly under water deficit, whereas activity of C 4 pathway enzymes was significantly enhanced, especially that of phosphoenolpyruvate carboxylase and NADP-malate dehydrogenase. Water deficit exerted lesser influence on spike photosynthesis in Pub. Ear organs exhibited delayed senescence. Accumulation of photosynthetic carbon assimilates in ear bracts occurred mainly during the early grain filling and photosynthates were transported in the middle of grain filling. C 4 pathway enzymes seem to play an important function in ear photosynthesis. We speculate that the high enzyme activity of the C 4 pathway and the increased capacity of photosynthetic carbon assimilate transport were the reasons for the drought tolerance characteristics of ears. ABSTRACT. Okra (Abelmoschus esculentus L.) is not only a nutrientrich vegetable but also an important medicinal herb. Inter-simple sequence repeat (ISSR) markers were employed to investigate the genetic diversity and differentiation of 24 okra genotypes. In this study, the PCR products were separated by electrophoresis on 8% nondenaturing polyacrylamide gel and visualized by silver staining. The 22 ISSR primers produced 289 amplified DNA fragments, and 145 (50%) fragments were polymorphic. The 289 markers were used to construct the dendrogram based on the unweighted pair-group method with arithmetic average (UPGMA) cluster analysis. The dendrogram indicated that 24 okras were clustered into 4 geographically distinct groups. The average polymorphism information content (PIC) was 0.531929, which showed that the majority of primers were informative. The high values of allele frequency, genetic diversity, and heterozygosity showed that primer-sample combinations produced measurable fragments. The mean distances ranged from 0.045455 to
The oriental armyworm, Mythimna separata, is a serious agricultural pest in China. Seasonal and roundtrip migration has recently led to sudden, localized outbreaks and crop losses. To evaluate genetic differentiation between populations in eastern and western China and elucidate gene flow, the genetic structure of 20 natural populations from nine provinces was examined using seven microsatellite markers. The results indicated high genetic diversity. However, little to moderate (0 < F ST < 0.15) genetic differentiation was detected, and there was no correlation between genetic distance and geographical distance. Bayesian clustering analysis identified three groups whereas discriminant analysis of principal components identified ten clusters that were considered as two clear‐cut clusters and one admixed group. Gene flow occurred frequently in most population pairs, and an asymmetrical migration rate was detected in several pairwise population comparisons. The bottleneck test showed that few populations had experienced recent bottlenecks. Correspondingly, large‐scale and long‐distance migration of M. separata has caused low genetic differentiation and frequent gene exchange. Our findings are important for studying genetic evolution and help to improve predictions of M. separata outbreaks in China.
To clarify association between armyworm (Mythimna separata) damage level and the corn growth and weed occurrence, we investigated corn plant height, stem diameter and vigor as well as weed coverage and biomass. The investigations were conducted at three locations of Shaanxi Province which were suffered seriously from armyworm. Significant correlations were found between the parameters analyzed. At stunted corn growth and presence of plenty of weeds, the armyworm damage tended to be heavy; oppositely, when corn grew well and weed density were low, armyworm harm was the minimal. Therefore, corn growing status and weed density can significantly affect armyworm damage level. Our results imply that promoting corn growth and timely removal of weeds are conducive to reducing armyworm occurrence.
