In an attempt to develop a system for producing transformed plants from explants of Dendranthema grandiflora, the susceptibility of the cultivar Super White to various wild-type strains of Agrobacterium tumefaciens and A. rhizogenes was investigated. Tumour formation was not a reliable indicator of the ability of a related disarmed strain to mediate transformation. Following inoculation of explants with disarmed Agrobacterium strains, a number of shoots developed on selective media. However, none of these shoots were transformed. By co-cultivating stem internode explants with a mixed inoculum of wild-type and disarmed strains, it was possible to obtain a callus stably transformed with Agrobacterium carrying a disarmed T-DNA. Histological analysis of explants revealed that shoot regeneration initially occurred from the cells of the epidermis and subsequently from the cortex. However, the cells which were susceptible to T-DNA transfer were confined to the vascular tissue.

Восточноазиатский вид Chrysanthemum leiophyllum является источником адаптивных при-знаков в селекции Ch. × morifolium. Исследовали особенности введения в культуру in vitro и клонального микроразмножения Chrysanthemum leiophyllum. В качестве эксплантов использовали части побега с пазуш-ными почками. Разработан способ стерилизации побегов, который обеспечивает получение жизнеспособных эксплантов. При культивировании на среде Мурасиге-Скуга (MC), дополненной 0,5 мг/л 6-бензиламинопу-рина (БАП) и 30 г/л сахарозы, формировались укороченные и витрифицированные побеги, в базальной ча-сти которых развивалась каллусная ткань. Уменьшение содержания БАП до 0,2 мг/л способствовало росту побегов. Снижение концентрации сахарозы в два раза приводило к увеличению коэффициента размножения Ch. leiophyllum до 10,3, а при снижении концентрации макросолей коэффициент размножения достигал 11,7. Оптимальной для размножения побегов является среда с половинным содержанием макросолей в сочетании с 0,2 мг/л БАП. Интенсивное развитие корней отмечено на MC-среде с половинным содержанием макросо-лей в присутствии 0,5 мг/л индолилуксусной кислоты (ИУК) и 30 г/л сахарозы. На развитие корней в боль-шей степени влияли регуляторы роста, оптимальным регулятором роста на стадии укоренения является ИУК. Эффективность адаптации растений к почвенным условиям составила 98 %. Полученные in vitro растения нормально развивались, к концу первого года в открытом грунте образовали приземные розетки листьев, а на второй год вступили в генеративную фазу. The specifics of introduction of Ch. leiophyllum into an in vitro culture and its micropropagation were studied. Stem segments with axillary buds were used as explants. A technique for sterilization of stems to obtain viable explants was developed. Shorter and vitrified shoots, with the callus tissue developing in their basal part, formed on Murashige and Skoog (MS) medium supplemented with 0,5 mg/L 6-benzylaminopurine (BAP) and 30 g/L sucrose. A decrease in the BAP level to 0.2 mg/L promoted shoot growth. A two-fold decrease in the sucrose concentration raised the multiplication coefficient of Ch. leiophyllum shoots to 10,3, while a two-fold decrease in the macro-salts concentration raised the multiplication coefficient to11.7.
