Mitotic homologous recombination (HR) is a critical pathway for the accurate repair of DNA double strand breaks (DSBs) and broken replication forks. While generally error-free, HR can occur between misaligned sequences, resulting in deleterious sequence rearrangements that can contribute to cancer and aging. To learn more about the extent to which HR occurs in different tissues during the aging process, we used Fluorescent Yellow Direct Repeat (FYDR) mice in which an HR event in a transgene yields a fluorescent phenotype. Here, we show tissue-specific differences in the accumulation of recombinant cells with age. Unlike pancreas, which shows a dramatic 23-fold increase in recombinant cell frequency with age, skin shows no increase in vivo. In vitro studies indicate that juvenile and aged primary fibroblasts are similarly able to undergo HR in response to endogenous and exogenous DNA damage. Therefore, the lack of recombinant cell accumulation in the skin is most likely not due to an inability to undergo de novo HR events. We propose that tissue-specific differences in the accumulation of recombinant cells with age result from differences in the ability of recombinant cells to persist and clonally expand within tissues.

Prihvaćeno u ožujku 2010.Mikronukleus (MN) test na limfocitima periferne krvi jedna je od najvažnijih metoda koje se primjenjuju u citogenetičkom nadzoru. Osnovni preduvjet za primjenu nekog testa u svrhu nadzora profesionalno izloženih populacija jest poznavanje normalnih vrijednosti promatranoga biološkog pokazatelja (biomarkera) u kontrolnoj populaciji. Baze podataka na razini opće populacije moraju se redovito obnavljati novim podacima. Cilj ovog istraživanja bio je utvrditi normalne i granične vrijednosti MN-testa na limfocitima periferne krvi 200 zdravih ispitanika obaju spolova iz opće populacije Republike Hrvatske te ispitati koji čimbenici pridonose spontanom nastanku MN. Na razini istražene populacije utvrđeno je prosječno (6,90±3,32) MN (medijan 7 MN), dok je raspon pojedinačnih vrijednosti iznosio 0 do 18 MN u 1000 binuklearnih stanica. Gornja granična vrijednost dobivena izračunavanjem 95. percentila za cjelokupnu promatranu populaciju iznosi 12,5 MN na 1000 limfocita. Utvrđeno je da na spontani nastanak MN utječu spol, dob i navika pušenja. Žene u prosjeku imaju više vrijednosti svih parametara MN-testa od muškaraca, a u njih je bio i naglašeniji porast vrijednosti citogenetičkog nalaza zbog navike pušenja. Kako su literaturni podaci o utjecaju pušenja cigareta na nastanak MN kontradiktorni, planiran je nastavak istraživanja radi razjašnjavanja utjecaja dnevno utrošenog broja cigareta i ukupnog trajanja pušačkog staža na vrijednosti parametara MN-testa. Usporedba rezultata s literaturnim podacima potvrdila je da su dobivene vrijednosti u skladu s vrijednostima MN-testa zabilježenim na općoj populaciji u drugim svjetskim laboratorijima. Normalne i granične vrijednosti MN-testa utvrđene u ovome istraživanju poslužit će kao osnova za usporedbu i tumačenje nalaza MN-testa u ispitanika izloženih populacija te daljnju nadogradnju laboratorijske baze podataka. KLJUČNE RIJEČI: citogenetički nadzor, dob, nestabilnost genoma, pušenje cigareta, rizik, spol, spontani nastanak mikronukleusaBiološki nadzor (engl. biomonitoring) važan je dio zdravstvenog nadzora nad ispitanicima koji su profesionalno izloženi različitim fizikalnim i kemijskim mutagenima i/ili kancerogenima. Zasniva se na mjerenju (ili utvrđivanju učestalosti) različitih bioloških pokazatelja koji upućuju na najranije, još popravljive biološke učinke, koji prethode pojavi zloćudnih i/ili drugih bolesti. Primjenom biljega izloženosti, učinka i sklonosti (engl. susceptibility) na razini proučavanih populacija možemo utvrditi kakve su razine izloženosti štetnostima na radnim mjestima, je li izloženost rezultirala mjerljivim učinkom (primjerice oštećenjem molekule DNA) te otkriti ispitanike s većom osjetljivosti genoma koji su pod povišenim * svi autori su jednakovrijedni Kopjar N, et al.

RESUMENLa integridad genética de la población humana se encuentra comprometida por la gran actividad industrial; por lo que es importante determinar qué se conoce como un nivel "aceptable" de daño genético y realizar ensayos de genotoxicidad de manera rutinaria en poblaciones de riesgo. Los micronúcleos son cuerpos citoplasmáticos de naturaleza nuclear, se corresponden con material genético no incorporado correctamente a las células hijas durante la división celular, reflejan aberraciones cromosómicas y se originan por roturas cromosómicas, por errores durante la replicación y posterior división celular del ADN y/o por la exposición a agentes genotóxicas. Existen factores capaces de influir o modificar el número de micronú-cleos presentes en una célula (edad, género, vitaminas, tratamientos médicos, exposición diaria a agentes genotóxicos, etc.).El ensayo citogenético para la detección de micronúcleos (CBMN: cytokinesis-block micronucleus) se basa en la utilización de un agente químico, denominado citocalasina-B capaz de impedir la citocinesis permitiendo la división nuclear proporcionando a las célu-las un aspecto de células binucleadas monodivididas. El recuento de micronúcleos se realiza sobre 1.000 células binucleadas y la muestra de partida puede variar aunque lo óptimo es el uso de linfocitos aislados de sangre periférica.El ensayo de micronúcleos está considerado como un ensayo práctico, universalmente validado y accesible tecnológicamente, útil para evaluar la inestabilidad genética inducida por agentes genotóxicos.Palabras clave.
