Normal and Cut-Off Values of the Cytokinesis-Block Micronucleus Assay on Peripheral Blood Lymphocytes in the Croatian General Population

Abstract: Institut za medicinska istraživanja i medicinu rada, Zagreb, HrvatskaPrimljeno u veljači 2010. Prihvaćeno u ožujku 2010.Mikronukleus (MN) test na limfocitima periferne krvi jedna je od najvažnijih metoda koje se primjenjuju u citogenetičkom nadzoru. Osnovni preduvjet za primjenu nekog testa u svrhu nadzora profesionalno izloženih populacija jest poznavanje normalnih vrijednosti promatranoga biološkog pokazatelja (biomarkera) u kontrolnoj populaciji. Baze podataka na razini opće populacije moraju se redovito ob… Show more

“…Many studies indicate that the MN frequency in humans progressively increases with age from early adulthood through the rest of life [25,30,[60][61][62][63][64][65]. In contrast, there are some studies that do not find such an increase [66].…”

“…Pareto charts of t-values for the influence of gender and age on alkaline comet-assay parameters (tail length, tail intensity and tail moment) determined in healthy children. Over the past years our laboratory has accumulated many data on cytogenetic biomarkers of general [27][28][29][30] and various exposed human populations [31][32][33][34][35][36]. Nevertheless, these biomarkers have to be re-evaluated occasionally, especially in terms of the general population, in order to establish the upper level of normal variability so that positive effects of exposure can be more easily recognized.…”

“…The micronucleus test was performed following the guidelines of Fenech and Morley [9] with minor modifications [10]. Blood (0.5 mL) was added to RPMI 1640 medium (Gibco, USA) supplemented with foetal bovine serum (Gibco) and phytohaemagglutinin (Remel Europe Ltd., UK) and incubated at 37 • C and 5% CO 2 for 72 h. After 44 h of culture, cytochalasin-B (Sigma, USA) was added (final concentration 6 g/mL), in order to prevent cytokinesis.…”

“…Studije su pokazale da među polovima postoje različite frekvence mikronukleusa u limfocitima periferne krv i da je frekvence kod žena viša u odnosu na muškarce, što se tumači gubitkom jednog od X hromozoma putem mikronukleusa (Bonassi et al, 2001;Kopjar et al, 2010).…”
