Seryl-tRNA Synthetase Shows a Noncanonical Activity of Upregulating Laccase Transcription in Trametes hirsuta AH28-2 Exposed to Copper Ion

Abstract: Seryl-tRNA synthetase is well known for the attachment of serine to the corresponding cognate tRNA during protein translation. In contrast, its functions beyond translation in microorganisms are underexplored.

“…S4). Despite the lack of change in ThserRS expression levels, serine in C. unicolor 87613 did show the same negative impact on laccase production as TherRS reported in T. hisuta AH28-2 [51]. Hence, it remains a possible way to negatively influence fungal laccase production through the interaction between serine and TherRS (or other TFs).…”

“…ferulae [50]. Moreover, Gan and collaborators uncovered a novel role for seryl-tRNA synthetase (TherRS) as a TF in the repression of laccase production through its interaction with the promoter region of the laccase gene in T. hisuta AH28-2 [51]. These findings suggest a potential mechanism by which serine might regulate laccase production through interaction with TFs, such as TherRS.…”

“…The C-terminal extension specific for vertebrate SerRSs, the unique domain UNE-S, harbors robust NLS that redirects the protein from the cytosol into the nucleus, where it exhibits non-canonical functions unrelated to translation [63]. Interestingly, the SerRS from the fungus Trametes hirsuta also shows nuclear localization and acts as a transcriptional repressor [64]. An evolutionary addition to higher eukaryote cytosolic SerRSs are two insertions (Figure 1).…”
