Serum miR-300 as a diagnostic and prognostic biomarker in osteosarcoma

Abstract: In order to determine whether microRNA (miR)-300 is a diagnostic and prognostic biomarker in osteosarcoma, the miR-300 levels in serum of 114 osteosarcoma patients and 114 healthy controls were compared, followed by serum analysis of the differences between the pre-operative and post-operative sera of these osteosarcoma patients. It was observed that the concentration levels of miR-300 in the serum of osteosarcoma patients was significantly higher than those in the serum of healthy controls (P<0.01). Furthermo… Show more

“…For example, downregulation of miR-95-3p was found in osteosarcoma patients compared to healthy controls, and low miR-95-3p expression was positively correlated with clinical stage and metastasis, as well as shorter survival and disease-free survival, indicating miR-95-3p served as a tumor suppressor in osteosarcoma [ 10 ]. Conversely, Liu et al showed miR-300 levels in osteosarcoma cases were significantly higher than those in healthy controls, and aberrantly elevated miR-300 expression was associated with worse prognosis of patients, suggesting an oncogenic role of miR-300 [ 11 ].…”

“…During tumor initiation and progression, miRNAs are found to involve in diverse biological processes, such as cell proliferation, invasion, metastasis and angiogenesis [ 8 , 9 ]. Several miRNAs, such as miR-95-3p [ 10 ], miR-300 [ 11 ], miR-101, are found to be aberrantly expressed in osteosarcoma.…”

“…Similarly to miR-199a-5p, serum miR-300 levels decreased in OS patients following curative surgery. High levels of miR-300 were detected in advanced clinical stages and in metastasis, and indicated as an independent predictor factor for poor overall survival and progression free-survival rate in OS [ 69 ]. Of note, the authors underscored an important limitation of using miR-300 as a single biomarker for early detection and monitoring of OS: in fact, circulating miR-300 has been involved in the pathogenesis of other cancers, such as glioma and head and neck squamous cell carcinoma [ 70 , 71 ].…”
