Circulating biomarkers in osteosarcoma: new translational tools for diagnosis and treatment

Abstract: Osteosarcoma (OS) is a rare primary malignant bone tumour arising from primitive bone-forming mesenchymal cells, with high incidence in children and young adults, accounting for approximately 60% of all malignant bone tumours. Currently, long-term disease-free survival can be achieved by surgical treatment plus chemotherapy in approximately 60% of patients with localized extremity disease, and in 20–30% of patients with metastatic lung or bone disease. Diagnosis of primary lesions and recurrences is achieved b… Show more

“…A promising new approach to cancer diagnosis exploits circulating biomarkers that can be repeatedly and conveniently assessed with minimal complications. This is especially true of circulating exosomes, which contain a number of biomarkers specific to the originating cells . As a result, exosome analysis has garnered increasing attention owing to its novel features including the fact that exosomes function as reliable surrogates of parental cells and that they are both abundant and structurally stable.…”

“…Several studies have examined c-miRNAs in OS [50], but few studies have been conducted in other sarcoma subtypes. Here, we used next generation sequencing to compare c-miRNA profiles of four sarcoma subtypes: OS, SS, ES, and GIST.…”

“…Liquid biopsy may be highly advantageous, given the difficulty of recovering the bone sample for disease staging. In fact, it is a non-invasive and also time-saving approach; moreover, it may offer more precise and accurate information on early detection, therapeutic decisions, and response to therapy in OS [12,13].…”
