Restriction of food intake leads to immunologic and histomorphological changes in rats demonstrated by Moriyama. This study is planned to show how sleeve gastrectomy restricts food intake and its effects on biochemical liver function, immunologic and hematologic laboratory parameters, and histomorphological changes in rats. We used sleeve gastrectomy model on seven young adult female rats and compared study group with sham and control groups. Food consumption of rats was measured. All rats were sacrificed on the 50th day, and blood and tissue samples were collected. There was a significant low food intake in sleeve gastrectomy group (p=0.013). No differences were observed on hematologic, biochemical, and immunologic laboratory parameters between groups. Toxicity parameters in liver samples such as cytoplasmic atrophy, single-cell hepatocellular necrosis, and necrotic eosinophilic cells were significantly high in sleeve gastrectomy group (p=0.005). Histomorphological examination of the spleen and kidneys revealed significant changes in sleeve gastrectomy and sham groups compared with controls (p=0.004 and p=0.018, respectively). Although sleeve gastrectomy does not lead to alteration in hematologic, biochemical, and immunologic laboratory parameters, it causes decreased food consumption, which results in toxicological histomorphological changes in rat liver as well as some changes in kidney and spleen samples.

Effects of adiponectin on breast cancer cell growth and signaling

Obesity is a risk factor for postmenopausal breast cancer. Adiponectin/Acrp30 is lower in obese individuals and may be negatively regulating breast cancer growth. Here we determined that five breast cancer cell lines, MDA-MB-231, MDA-MB-361, MCF-7, T47D, and SK-BR-3, expressed one or both of the Acrp30 receptors. In addition, we found that the addition of Acrp30 to MCF-7, T47D, and SK-BR-3 cell lines inhibited growth. Oestrogen receptor (ER) positive MCF-7 and T47D cells were inhibited at lower Acrp30 concentrations than ER-negative SK-BR-3 cells. Growth inhibition may be related to apoptosis since PARP cleavage was increased by Acrp30 in the ER-positive cell lines. To investigate the role of ER in the response of breast cancer cells to Acrp30, we established the MDA-ERa7 cell line by insertion of ER-a into ER-a-negative MDA-MB-231 cells. This line readily formed tumours in athymic mice and was responsive to oestradiol in vivo. In vitro, MDA-ERa7 cells were growth inhibited by globular Acrp30 while the parental cells were not. This inhibition appeared to be due to blockage of JNK2 signalling. These results provide information on how obesity may influence breast cancer cell proliferation and establish a new model to examine interactions between ER and Acrp30.

Aim: To evaluate the effects of borax and sleeve gastrectomy on mRNA expression of antioxidant genes in substantia nigra tissue of obese rats. Methods:Obese rats were fed with a high-fat diet containing 40% additional fat to the diet. Rats were allocated into four groups in random, which were normal rats (Group I) (n=14), obese rats subjected to SG (Group II) (n=14), obese rats subjected to borax (Group III) (n=14), and obese rats subjected to SG and borax (Group IV) (n=14). Catalase, superoxide dismutase (SOD) and glutathione S-transferase (GST) gene expressions were determined by polymerase chain reaction, real-time polymerase chain reaction (RT-qPCR) and western blotting.Results: When normal rats (Group I), and obese rats subjected to SG (Group II) were compared, a decrease in expressions of catalase, SOD and GST genes was observed in Group II. When obese rats subjected to borax (Group III) were compared with Group I and Group II, a decrease in expressions of catalase, SOD and GST genes was observed in Group III. This phenomenon demonstrates that borax and SG both decrease expressions of catalase, SOD and GST genes. Furthermore, the most significant decrease in expressions of catalase, SOD and GST genes was observed in obese rats subjected to SG and borax (Group IV) when compared to other three study groups. Conclusion:The borax decreases molecular obesity and consequently increases the expressions of Catalase, SOD and GST genes. These data show decrease of Catalase, SOD and GST genes in the substantia nigra tissue of obese rats, consistent with the possibility that these changes may contribute to disease pathogenesis.

Abstract: Unlike after diet or gastric restrictive surgery, BPD-DS is associated with markedly suppressed ghrelin levels, possibly contributing to the weight-reducing effect of this operation. Sleeve gastrectomy seems to be the main cause of this reduction.
