Serum concentrations of TNF-α and soluble TNF-α receptors in obesity

Abstract: OBJECTIVE: To study whether weight reducing treatment modulates serum concentration of TNF-a a and two soluble TNF-a a receptors in obese subjects. SUBJECTS AND MEASUREMENTS: Serum concentrations of TNF-a a and two soluble receptors (sTNF-R1, sTNF-R2), plasma glucose, insulin, total cholesterol, HDL-cholesterol and triglicerides were measured in 27 obese subjects (age 48 AE 12 y, body mass index (BMI): 36 AE 6 kgam 2 ) before and after 3 months weight reducing treatment consisted of a diet & 1000 kcaladay and … Show more

“…This is noteworthy as decreased TNFRSF1B protein expression has been linked to increases in TNFα-induced apoptosis in cells that are crucial to immuno-regulation and inflammatory responses such as lymphocytes [32]. Considering our findings, in conjunction with findings that describe (a) the ability of chronic aerobic exercise to significantly reduce TNFα mRNA expression [33], (b) the inverse relationship between serum levels of TNFα and TNF receptors [34], and (c) the inverse relationship between TNFα-induced apoptosis and TNFRSF1B protein expression [32], we propose that HIIT favourably influences TNFα activity, as evidenced by an increase in the expression of Tnfrsf1b mRNA. Furthermore, based on our data, we propose the ability of exercise to influence TNFα activity is intensity-dependant as HIIT resulted in a significant up-regulation of Tnfrsf1b mRNA expression, compared to LIT (p=0.047).…”

“…103 TNF-a is a strong inducer of lipolysis 104 and data from rodents and humans support a role in the development of insulin resistance. [105][106][107] The latter, however, has been challenged by others 108 and although TNF-a levels are increased in obesity, 109,110 there is little direct release from abdominal subcutaneous adipose tissue in vivo. 111 Interestingly, in a similar set of studies it was found that abdominal subcutaneous adipose tissue releases the soluble TNF receptor type 1 and that its levels correlate with BMI.…”
