SERS-Based Colloidal Aptasensors for Quantitative Determination of Influenza Virus

Abstract: Development of sensitive techniques for rapid detection of viruses is on a high demand. Surface-enhanced Raman spectroscopy (SERS) is an appropriate tool for new techniques due to its high sensitivity. DNA aptamers are short structured oligonucleotides that can provide specificity for SERS biosensors. Existing SERS-based aptasensors for rapid virus detection had several disadvantages. Some of them lacked possibility of quantitative determination, while others had sophisticated and expensive implementation. In … Show more

“…Next, 0.03% NaN 3 was added and viruses were stored at +4°C. Hemagglutinating activity and concentrations of the virus in virions/mL were determined as described in [ 15 ]. The design of our study was approved by the Ethics Committee of the Chumakov Center (Approval no.…”

“…A monotonous drop in the SERS signal upon increasing the concentration of virus particles was observed on colloidal silver particles when reproducing the sandwich technique for influenza A viruses. The range of determined concentrations was 2 × 10 5 –2 × 10 6 particles/mL [ 15 ]. when detecting the SARS-CoV-2 coronavirus, there was a monotonously rising dependence of signal intensity on the concentration of the virus with a 5.5 × 10 4 –1.4 × 10 6 TCID50/mL range of detection [ 16 ].…”

“…Aptamers are single-stranded oligonucleotides with nucleic acid sequences forming a 3D structure, which can specifically bind a target molecule [71]. Gribanyov et al used an aptamer against hemagglutinin (HA) for SERS-based detection of the influenza A virus [72]. Chen et al also introduced an aptamer-based strategy for detection of Influenza A targeting HA (Figure 9) [73].…”

“…Two types of DNA aptamers were incubated in aqueous AgNP environment (used as the base of the sensor): aptamers RHA0385 and BV42, with an affinity for a variety of HA antigens of influenza A strains. These aptasensors detected influenza A virus particles and showed an innovation of quantitative detection for SERS-based assays, proving that DNA–RNA interactions (aptamer and viral particles, respectively) are more effective for this diagnostic analysis when compared to DNA–DNA interactions [ 47 ].…”
