Leishmania mexicana (L. mexicana) is a causal agent of cutaneous leishmaniasis (CL), a “Neglected disease”, for which the search for new drugs is a priority. Benzimidazole is a scaffold used to develop antiparasitic drugs; therefore, it is interesting molecule against L. mexicana. In this work, a ligand-based virtual screening (LBVS) of the ZINC15 database was performed. Subsequently, molecular docking was used to predict the compounds with potential binding at the dimer interface of triosephosphate isomerase (TIM) of L. mexicana (LmTIM). Compounds were selected on binding patterns, cost, and commercial availability for in vitro assays against L. mexicana blood promastigotes. The compounds were analyzed by molecular dynamics simulation on LmTIM and its homologous human TIM. Finally, the physicochemical and pharmacokinetic properties were determined in silico. A total of 175 molecules with docking scores between −10.8 and −9.0 Kcal/mol were obtained. Compound E2 showed the best leishmanicidal activity (IC50 = 4.04 µM) with a value similar to the reference drug pentamidine (IC50 = 2.23 µM). Molecular dynamics analysis predicted low affinity for human TIM. Furthermore, the pharmacokinetic and toxicological properties of the compounds were suitable for developing new leishmanicidal agents.

La leishmaniasis y la enfermedad de Chagas son causadas por protozoarios que comparten características filogenéticas y eco-epidemiológicas comunes, lo cual provoca dificultad en su identificación y diagnóstico. El objetivo de este trabajo fue analizar la información disponible sobre las técnicas de identificación de T. cruzi, de Leishmania spp. y la coinfección con ambos parásitos, así como su impacto en la obtención de datos eco-epidemiológicos. Se realizó una búsqueda sistemática de técnicas de identificación para T. cruzi, Leishmania spp. o ambos. Los estudios se clasificaron por agente etiológico, tipo de publicación, técnica utilizada y hospedero. Se realizó una prueba de X2 para evaluar la relación entre el parásito y la técnica utilizada. También se analizó la relación entre los trabajos publicados por año, la técnica diagnóstica y el parásito. De 138 trabajos analizados, se determinó que no existe un “estándar de oro” para el diagnóstico de estas parasitosis y la coinfección. Por tanto, se requiere la estandarización de protocolos que incrementen la sensibilidad y especificidad, así como el uso de al menos 2 técnicas en la identificación. Es imprescindible estudiar la prevalencia e incidencia de estas parasitosis en la fauna silvestre y la población humana bajo el contexto de cambio climático.

Technological advances in the serological diagnosis of Chagas disease in dogs and cats: a systematic review

Background Chagas disease (CD) is caused by Trypanosoma cruzi, which is transmitted mainly through the feces/urine of infected triatomine bugs. The acute phase lasts 2–3 months and is characterized by high parasitemia and nonspecific symptoms, whereas the lifelong chronic phase features symptoms affecting the heart and/or digestive tract occurring in 30–40% of infected individuals. As in humans, cardiac abnormalities are observed in T. cruzi-infected dogs and cats. We reviewed the technological advances in the serological diagnosis of CD in dogs and cats. Methods A review of the published literature during the last 54 years (1968–2022) on the epidemiology, clinical features, diagnosis, treatment and prevention of CD in dogs and cats was conducted. Results Using predefined eligibility criteria for a search of the published literature, we retrieved and screened 436 publications. Of these, 84 original studies were considered for inclusion in this review. Dogs and cats are considered as sentinels, potentially indicating an active T. cruzi transmission and thus the risk for human infection. Although dogs and cats are reputed to be important for maintaining the T. cruzi domestic transmission cycle, there are no commercial tests to detect past or active infections in these animals. Most published research on CD in dogs and cats have used in-house serological tests prepared with native and/or full-length recombinant antigens, resulting in variable diagnostic performance. In recent years, chimeric antigens have been used to improve the diagnosis of chronic CD in humans with encouraging results. Some of them have high performance values (> 95%) and extremely low cross-reactivity rates for Leishmania spp., especially the antigens IBMP-8.1 to IBMP-8.4. The diagnostic performance of IBMP antigens was also investigated in dogs, showing high diagnostic performance with negligible cross-reactivity with anti-Leishmania infantum antibodies. Conclusions The development of a commercial immunodiagnostic tool to identify past or active T. cruzi infections in dogs and cats is urgently needed. The use of chimeric recombinant T. cruzi antigens may help to fill this gap and is discussed in this review. Graphical Abstract
