Feline herpesvirus type 1 (FHV-1) causes respiratory and ocular disease in cats. Although isolates of FHV-1 circulating in cats have been reported worldwide, Korean FHV-1 isolates and their features have not been reported thus far. We aimed to investigate the biological and molecular characterization of two FHV-1 isolates based on the nucleotide sequence of thymidine kinase (TK) and glycoprotein B (gB) gene. In total, 48 samples from 12 cats were prepared for virus isolation. For the diagnosis, virus isolation, indirect fluorescence assay (IFA), electron microscopy (EM), and polymerase chain reaction (PCR) and for the molecular characterization, cloning and sequencing were used. Based on many methods such as virus isolation with specific cytopathic effects, IFA, EM, and PCR, two isolates were confirmed as FHV-1 and they showed the highest viral titer (10 8.3 to 10 8.5 TCID 50 /mL) in the Crandell-Rees Feline Kidney cells at 48 h after inoculation, but did not grow in MDCK and Vero cells. The nucleotide and amino acid sequences of the full TK and gB gene of FHV191071 and FHV191072 isolates were determined and compared with those of other herpesvirus strains. Two isolates possessed the same nucleotide sequences belonging to FHV-1 group and had the highest similarity (99.9%) with the KANS-02 strain, which was isolated from shelter in USA in 2016. Two isolates were confirmed as FHV-1 and they will be a useful basic resource for evaluating current FHV-1 vaccine and developing diagnostic tools.

ABSTRACT. A serosurvey of feline herpesvirus type 1 (FHV-1), feline calicivirus (FCV), and feline parvovirus (FPV) in cats from Ho Chi Minh City area in southern Vietnam was conducted in December 1998, and we compared the results with our previous results in northern Vietnam (Hanoi area). The positive rate of FHV and FCV in domestic cats were 44% and 74%, respectively. They were rather higher than those in Hanoi area, while the seropositivity of FPV (44%) was similar to that in Hanoi area. In leopard cats, the positive rate of FPV was high (3/4) and it indicated that FPV was prevailing in leopard cats in Vietnam.-KEY WORDS: feline calicivirus, feline herpesvirus type 1, feline parvovirus.J. Previously we conducted a serosurvey for the viruses in domestic and leopard cats from Hanoi area in northern Vietnam in 1997 [8, 10]. In the present study, we expanded the study and collected samples of cats from Ho Chi Minh City (HCM) area in southern Vietnam and compared the results with our previous reports in northern Vietnam. As a consequence, we found that prevalence of the viruses was different between the areas. A total of 54 blood samples were collected from 50 domestic cats (Felis catus) and 4 leopard cats (Felis bengalensis) in December 1998. Most of the domestic cats were less than 4 years old. In addition, all the cats are considered to be unvaccinated for prophylaxis of viral infections because the vaccines are not commercially sold. As for the leopard cats, one cat from the Saigon ZooBotanical Garderns, one cat was household, two cats were captured in the vicinity of HCM. The cats were immobilized by an intramuscular injection with 20 mg/kg of Ketamine before sampling. The blood samples, from which plasmas were isolated, were collected by adding heparin for anticoagulation. The antibodies against FHV-1, FCV and FPV were tested by the indirect immunofluorescence assay (IFA) as described previously [8]. Plasma samples were screened at a dilution of 1:50 in the IFA.The results of this survey together with our previous reports [8,10] were summarized in Table 1. In Hanoi area, a positive rate for FHV-1 antibody was only 1.4%. The positive rates of FCV and FPV were 39 and 54%, respectively. On the contrary, in HCM area, the positive rate of FHV-1 antibody was 44%, which is by far higher than in Hanoi area. In addition, we found that the positive rate in male cats was three times higher than that in female cats. The positive rate of FCV antibody was 74% which was two times higher than that in Hanoi area. Unlike the FHV-1 infection, in FCV positive rates, we could not find a

ABSTRACT. Feline herpesvirus type 1 (FHV-1) is a causative agent of feline viral rhinotracheitis and belongs to the subfamily Alphaherpesvirinae of the family Herpesviridae. Since first isolated in 1958 by Crandell and Maurer, FHV-1 is distributed worldwide and is the most clinically significant agent for respiratory infections in cats. In this review, we describe the recent findings with properties and functions of FHV-1 glycoproteins, especially hemagglutinins. -KEY WORDS: feline herpesvirus type 1, glycoprotein, hemagglutinin.J. Vet. Med. Sci. 60(8): 881-888, 1998 negative staining of the virions [16,52,54,88] or ultrathin sectioning of FHV-1-infected cells [11,54], mature virus particles are 120-180 nm in diameter, with various morphologies in shapes and sizes of the envelope surrounding the hexagonal outline of the capsid. The genome of FHV-1 consists of approximately 126-134 kbp. It has a G+C content of approximately 50%. The genome is divided into two unique portions, the unique long (U L ) and unique short (U S ) regions, by inverted repeat segments (IR) (Fig. 1) [24,70]. The U L is 99-104 kbp in size and is composed of unique DNA. The adjacent short segment is approximately 27-30 kbp in size and contains a central portion of Us which is approximately 8-9 kbp in size. The Us region is bounded by inverted repeat sequences of 7-11 kbp.We have already introduced review articles on recombinant viral vector vaccines for the veterinary use [97] and molecular interactions between retroviruses and herpesviruses [30]. Therefore, this review mainly focuses on some of the recent developments regarding properties and functions of FHV-1 glycoproteins including hemagglutinin.
