Sequence related amplified polymorphism (SRAP) analysis for genetic diversity and micronutrient content among gene pools in mungbean [Vigna radiata (L.) Wilczek]

Abstract: Vigna radiata (L.) Wilczek, commonly called mungbean is an important pulse crop. Commercial cultivars contain low levels of iron and zinc and it is important to assess genetic variability in the available germplasm for improving micronutrient content in commercial cultivars. The present study was undertaken to study molecular diversity using Sequence-related amplified polymorphism (SRAP) among 21 Vigna radiata genotypes. Twenty nine SRAP primer combinations produced a total of 121 amplified bands which were po… Show more

“…rice, foxtail millet, pearl millet, melons, bottle gourd, Morinda tomentosa, proso millet etc. (Saini et al 2004;Kumar et al 2006;Collard and Mackill 2009;Luo et al 2010;Kumari et al 2011;Yildiz et al 2011;Aneja et al 2013;Arya et al 2014;Bhawna et al 2014;Trivedi et al 2015). The information generated from the present work will provide an estimate of the range of overall genetic variation of kodo millet existing in Indian sub-continent for designing suitable strategies for collection and conservation of germplasm for its enhancement and utilization.…”

“…In present study ML-776 (high Fe and Zn) and Satya (low Fe and Zn) genotypes showed reasonable diversity which are from different sub group that may be exploited or selecting parents for breeding programmes. In previous study genotype ML-776 also reported as high iron and zinc and Satya as low iron and zinc variety based on RAPD and SRAP analysis (Aneja et al, 2013).…”

“…A polymerase chain reaction (PCR) was carried out in 20 μL reaction volumes containing 150 ng of DNA, 1x buffer (150 mM Tris-HCl, pH 8.75 at 25 °C, 500 mM KCl, 20 mM MgCl2, 1% Triton X-100), 200 μM of each deoxyribonucleotide triphosphate (dNTPs), 0.6 μM of each forward and reverse primer, and 1U of Taq DNA polymerase (Vivantis, Selangor Darul Ehsan, Malaysia). The PCR conditions according to Aneja et al (2013) were followed. T100 TM Thermal Cycler (Bio-rad Laboratory, Inc., California, USA) was used for DNA amplification.…”

“…Because of its simplicity, high reproducibility and because no prior genome sequence is needed, this method has become attractive for breeding programs. Aneja et al (2013) and Alghamdi et al (2017) found that all SRAP primer combinations exhibited 100% polymorphism, thereby being useful in genetic assessment amongst mungbean accessions, as well as other Vigna spp. The objectives of this study were: i) to evaluate agronomic and photosynthetic performance of 23 mungbean and four blackgram genotypes and ii) to estimate their genetic diversity and relationships using SRAP markers.…”
