ABSTRACT:In this study, central composite design (CCD) was used for optimization of high performance liquid chromatographic (HPLC) method for simultaneous analysis of curcumin (CUR) and demethoxycurcumin (DMC) in tablets containing Curcuma extract. Separation of CUR and DMC was performed using X-Bridge C18 column (250 x 4.6 mm i.d; 5 µm). Four factors that were investigated include the concentration of acetic acid (X1), ratio of acetic acid (X2), flow rate of mobile phase (X3) and column temperature (X4). Based on responses obtained (retention time, peak area, resolution and tailing factor), the optimum condition selected was X1 = 3.00%, X2 = 51%, X3 = 1.05 mL/min and X4 = 45oC. This HPLC condition was validated by assessing several validation parameters including system suitability test, selectivity, linearity, precision, accuracy and robustness according to International Conference Harmonization (ICH). All validation parameters meet the acceptance criteria set by ICH. The validated method was successfully used for analysis of CUR and DMC in tablets containing Curcuma extract. CCD was effective means in optimization of HPLC for analysis of CUR and DMC in pharmaceutical formulation.

A survey of leaf spot disease on durian caused by Phomopsis durionis was conducted in durian growing areas in eastern Thailand, Chanthaburi and Trat provinces. It was found that lesions with yellow halos on both mature and young leaves are variable in sizes (1-10 mm in diameter). In this study, nine morphologically distinct isolates of Phomopsis were obtained from durian leaf spots. Some of them produced small number of pycnidia on potato dextrose agar a er incubation for 30 days. Artifi cial inoculation on wounded leaves of durian seedlings, resulted in the production of browning areas with yellow halos and pycnidium formation at 13 days and 20 days a er inoculation, respectively. Furthermore, red-brown spots with yellow halos on leaf tissues were observed at 32 days a er inoculation. High density of Phomopsis was observed in durian symptomless leaves and fl owers indicated the latent infection of the pathogen in the fi elds. Interestingly, crude extract of durian leaf with preformed substances demonstrated inhibition of spore germination and germ tube growth of the pathogen, Phomopsis sp., on water agar. In addition, bioassay on TLC plate displayed inhibition zone of growth of the fungus, Cladosporium oxysporum at retention factor (R f ) of 0.29-0.88. This indicates that preformed substances in leaf tissues might act as compounds aff ecting latent period of pathogen.

In order to enhance the bioavailability of curcumin its conjugates with piperic acid and glycine were synthesized by esterifying the 4 and 4' phenolic hydroxyls, the sites of metabolic conjugation. Antiproliferative and apoptotic efficacy of synthesized conjugates was investigated in MCF-7 and MDA-MB-231 cell lines. IC 50 values of di-O-glycinoyl (CDG) and di-O-piperoyl (CDP) esters of curcumin were found to be comparable with that of curcumin. Both conjugates induced chromatin condensation fragmentation and apoptotic body formation. CDP exposure to MCF-7 cells induced apoptosis initiating loss of mitochondrial membrane potential (∆ψm) followed by inhibition of translocation of transcription factor NF-kB and release of Cytochrome-C. Reactive oxygen species (ROS) production was evaluated by fluorescent activated cell sorter. Change in ratio of Bcl2/ Bclxl was observed, suggesting permeablization of mitochondrial membrane leading to the release of AIF, Smac and other apoptogenic molecules. DNA fragmentation as a hallmark for apoptosis was monitored by TUNEL as well as agrose gel electrophoresis. Thus, it was proven that conjugation does not affect the therapeutic potential of parent molecule in vitro, while these could work in vivo as prodrugs with enhanced pharmacokinetic profile. Pharmacokinetics of these molecules under in vivo conditions is a further scope of this study.
