Aflatoxin B (AFB ), one of the most toxic mycotoxins, is classified as a group I carcinogen and ubiquitous in various foods and agriproducts. Thus, accurate and sensitive determination of AFB is of great significance to meet the criteria of food safety. Direct detection of AFB is difficult by monoclonal antibody (mAb) with large molecular size (≈150 kD) since the target is too small to produce a detectable signal change. Herein, by combining the electrochemical properties of nanomaterials and the advantages of nanobodies, we developed a direct, highly selective and sensitive electrochemical immunosensor for small molecule detection. The proposed immunosensor had a wide calibration range of 0.01 to 100 ng mL and a low detection limit of 3.3 pg mL (S/N=3). Compared with the immunosensor prepared with mAb which was applied in the typical indirect immunoassay, the immunosensor in this work possessed two orders of magnitudes wider linear range and 10-fold more sensitivity. The as-obtained immunosensor was further successfully applied for sensing AFB in real samples. This proposed assay would provide a simple, highly sensitive and selective approach for the direct immunoassay of small molecule AFB , and is extendable to the development of direct immunosensing systems for other small molecules detection by coupling nanocarbon and nanobody.

The current study reveals anti-cancer properties of bio-functionalized silver nanoparticle (AgNPs) fabricated by Juniperus chinensis leaf extracts due to it’s easy, low cost, biological activity, eco-friendly and lower side effects. The characteristics of AgNPs were determined by scanning electron microscopy [1], transmission electron microscopy (TEM), UV-visible spectroscopy, Fourier transform infrared (FTIR) spectroscopy, X-ray diffraction, Dynamic light scattering (DLS), Zeta potential and X-ray spectroscopy (EDX). Further, this study highlights the cellular and molecular mechanisms of AgNPs involves in anti-proliferative and apoptotic effect on human lung cancer cells (A549) and compared to commercial drug cisplatin by various biological methods such as MTT, flow cytometery analysis, gene expression patterns, migration and invasion inhibition, ROS and caspase production and cell staining. The size of AgNPs fabricated in this study was 12.96 with spherical shape and negative zeta potential. Up-regulation of capase 3,9 and p53, Annexin V-FITC/PI, DAPI staining, ROS production indicated remarkable apoptotic effect of AgNPs than cisplatin. Also down-regulation of MMP2/MMP9 scratch and matrigel assays revealed anti-metastatic properties of AgNPs.cell cycle analysis and down regulation of cyclin D1 showed cancer cell cessation in the G0/G1 phase. Overall results in current experiment revealed AgNPs synthetized by biogreen method anti-metastasis and anti-proliferation effect on lung cancer cells comparison to cicplatin drug and also had lower side effect on normal cell line.

AlN thin film bulk acoustic resonators (FBARs) with a resonant frequency of $575 MHz have been fabricated to function as an epithelial tumor marker mucin 1 (MUC1) biosensor. Streptavidin was assembled on the sensitive area of FBAR. After the recognition between aptamers-AuNP conjugates and MUC1, biotin, along with the conjugates, was captured by streptavidin onto the surface of FBARs. Therefore, the target MUC1 could be sensitively detected. This biosensor exhibited a good linear relationship between the frequency shifts and the concentrations of MUC1 ranging from 30 to 500 nM, which indicated the sensitivity is about 818.6 Hz nM À1 . The frequency shift remained relatively stable when the concentration of MUC1 was greater than 500 nM since the binding between MUC1 and aptamers-AuNP conjugates reached saturation. The selectivity experiment demonstrated that this biosensor can precisely detect MUC1 with good specificity. The positive results suggest that FBAR is an attractive alternative to a new approach for the detection of MUC1.
