Kisspeptin acts as an upstream regulator of the hypothalamus-pituitary-gonad axis, which is one of the main regulatory systems for mammalian reproduction. Kiss1 and its receptor Kiss1r (also known as G protein-coupled receptor 54 (Gpr54)) are expressed in various organs, but their functions are not well understood. The purpose of this study was to investigate the expression profiles and functions of kisspeptin and KISS1R in the reproductive tissues of imprinting control region mice. To identify the expression pattern and location of kisspeptin and KISS1R in gonads, testes and ovarian tissues were examined by immunohistochemical or immunofluorescent staining. Kisspeptin and KISS1R were expressed primarily in Leydig cells and seminiferous tubules respectively. KISS1R was specifically localized in the acrosomal region of spermatids and mature spermatozoa. Kisspeptin, but not KISS1R, was expressed in the cumulus-oocyte complex and oviductal epithelium of ovarian and oviductal tissues. The sperm intracellular calcium concentrations significantly increased in response to treatment with kisspeptin 10 in Fluo-4-loaded sperm. The IVF rates decreased after treatment of sperm with the kisspeptin antagonist peptide 234. These results suggest that kisspeptin and KISS1R might be involved in the fertilization process in the female reproductive tract. In summary, this study indicates that kisspeptin and KISS1R are expressed in female and male gametes, respectively, and in mouse reproductive tissues. These data strongly suggest that the kisspeptin system could regulate mammalian fertilization and reproduction.

The similarities and differences of small RNAs in seminal plasma, epididymal sperm and ejaculated sperm remain largely undefined. We conducted a systematic comparative analysis of small RNA profiles in pig ejaculated sperm, epididymal sperm and seminal plasma and found that the diversity distribution of small RNA species was generally similar, whereas the abundance of small RNAs is dramatically different across the three libraries; miRNAs and small RNAs derived from rRNA, tRNA, small nuclear RNA, 7SK RNA, NRON RNA and cis-regulatory RNA were enriched in the three libraries, but piRNA was absent. A large population of small RNAs from ejaculated sperm are ejaculated sperm specific, and only 8-30% of small RNAs overlapped with those of epididymal sperm or seminal plasma and a small proportion (5-18%) of small RNAs were shared in the three libraries, suggesting that, in addition to the testes, sperm RNAs may also originate from seminal plasma, epididymis as well as other resources. Most miRNAs were co-distributed but differentially expressed across the three libraries, with epididymal sperm exhibiting the highest abundance, followed by ejaculated sperm and seminal plasma. The prediction of target genes of the top 10 highly expressed miRNAs across the three libraries revealed that these miRNAs may be involved in spermatogenesis, zygote development and the interaction between the environment and animals. Our study provides the first description of the similarities and differences of small RNA profiles in ejaculated sperm, epididymal sperm and seminal plasma and indicates that sperm RNA may have origins other than the testes.Reproduction (2017) 153 [785][786][787][788][789][790][791][792][793][794][795][796]

Article HistoryThe anaerobic probiotic technology (ZAD) is a patented technique. The products are in two forms either liquid or powder. The powder products are coated so it can handle the pressure and the temperature of the extruders. They are fed as follows; 10 gm/head /day for large animals or 1 kg/ tone of fed (for ZADO). In case of Liquid (ZAD); it is fed 10 ml/head/day for large animals and 1.5 ml liter for poultry and rabbits. It decreases the aflatoxins less than 11% by inclusion rate of 7% of ZAD in 7 hours. They are helping the animals to cope with heat stress. Their ability to decrease the cholesterol and triglycerides are noticed by 21 and 19%, respectively. The birds and animal immunity is elevated too. Animal performances are improved in animals fed either form. Milk production increased in dairy cattle (1.5 kg/animal/day) and dairy buffaloes (1kg/head/day). The same trend was recorded for meat production in cattle, buffaloes, and sheep (220, 180 and 90 gm/head/day, respectively as an average daily gain). For poultry, it helped in decreasing the fattening period by 5 days. In fish, the final weight increased by 18%. For rabbits, it increased the milk production, decreased the mortality rate, increased the average daily gain, and improved all over physiological aspects of rabbits.Contribution/Originality: This study documents the impact of probiotics on the growth performances and nutritional aspects of dairy animals, poultry, rabbit, fish, and beef. The primary contribution of this review article is to analyze the influence of anaerobic probiotic technology on the performances and other physiological aspects of ruminant and non-ruminant animals.
