Self-Assembled DNA Nanopores That Span Lipid Bilayers

Abstract: DNA nanotechnology excels at rationally designing bottom-up structures that can functionally replicate naturally occurring proteins. Here we describe the design and generation of a stable DNA-based nanopore that structurally mimics the amphiphilic nature of protein pores and inserts into bilayers to support a steady transmembrane flow of ions. The pore carries an outer hydrophobic belt comprised of small chemical alkyl groups which mask the negatively charged oligonucleotide backbone. This modification overcom… Show more

“…[22] The AFM-derived length and width of 13.0 AE 2.7 nm and 6.9 AE 1.5 nm (fullwidth-at-half-maximum), respectively, were in good agreement with the theoretical dimensions (14 nm and 5.5 nm). The experimental dimensions reported here are smaller than of similarly sized pores of previous studies [18,19] because the present AFM read-out examined densely packed barrels (Figure 2 c) which is different to the isolated structures of the preceding reports. We investigated whether NP-EP interacts with lipid bilayers by monitoring the UV-melting profiles of the DNA.…”

“…DNA has been used to build a variety of nanoscale structures including rotaxanes, [1][2][3] catenenes, 4 reconfigurable three-dimensional objects, 5 biomimetic systems [6][7][8] and chemically fuelled molecular motors. [9][10][11] Proteins possess larger chemical diversity than nucleic acids and are, therefore, attractive building elements in nanotechnology.…”
