Selective Cd4+ T-Cell Depletion Does Not Prevent Graft-Versus-Host Disease1

Abstract: Donor-derived CD4+ T cells may play a role in the development of graft-versus-host disease (GVHD) and graft-versus-leukemia reaction after allogeneic bone marrow transplantation (BMT). Therefore, we evaluated the effect of CD4+ T-cell depletion on GVHD and graft-versus-leukemia reaction after HLA-matched BMT. CD4 depletion was performed using anti-CD4 monoclonal antibodies and immunomagnetic beads, initially in small-scale experiments on bone marrow and granulocyte colony-stimulating factor-mobilized periphera… Show more

“…Moreover, donor lymphocyte infusions depleted of CD8+ cells induce remissions of relapsed chronic myelogenous leukemia, with a low rate of severe aGvHD, supporting the hypothesis that CD8+ lymphocytes are important effectors of aGvHD [20]. In contrast, HLA-matched BMT depleted of CD4+ cells did not prevent aGvHD [21]. …”

“…Conflicting data have been reported on whether T-cell depletion reduces the incidence of GVHD [17,[22][23][24]. However, infusing < 10 5 T cells/kg is considered necessary to avoid significant GVHD in fully HLA-matched unrelated transplants; 3 ×10…”

“…In addition, when CD4 and CD8 T-cell populations were examined separately, no difference was found in the killing efficiency or in the IC 50 value (data not shown). This information is relevant because CD4 and CD8 donor lymphocytes contribute to GVHD, as shown by donor lymphocyte infusion protocols involving the infusion of either CD4-depleted 20 or CD8-depleted donor lymphocytes. 21 LVЈVFas-transduced T lymphocytes demonstrated normal viability in the absence of drug, indicating the absence of autotoxicity-signaling in the absence of added drug (data not shown).…”
