Selection of Genetic Markers to Determine Diversity in Theileria annulata Populations after Recombination

Abstract: ÖZAmaç: Theileria annulata popülasyonlarında rekombinasyon sonrası oluşacak genetik çeşitliliğin ve rekombinasyonun yoğun olduğu kromozomal bölgelerin tespit edilmesi amacıyla kullanılacak polimorfik mini ve mikrosatellite markerlerin belirlenmesidir. Yöntemler: Markerlerin belirlenmesi amacıyla Unipro UGENE programı kullanılmıştır. T. annulata'nın rastgele karıştırılmış genomik dizilimlere göre gerçek DNA diziliminde 10 kat fazla tandem tekrar (TR) belirleyen skor, eşik değer olarak belirlenmiştir. Tekrarlı m… Show more

“…A total of 23 polymorphic mini-and micro-satellite markers were used to genotype parasite isolates, as described previously (Bilgic et al, 2017;Weir et al, 2007). These included 14 microsatellites (Tmsc 1,31,33,37,45,48,68,75,77,86 and TS 5,9,12,16) and nine minisatellites (MSC 8,19,and TS 6,8,15,20,25,31) distributed across each of the four chromosomes of the T. annulata genome.…”

The reagents required and thermocycler conditions selected for PCR amplification have been previously described (Bilgic et al, 2017;Weir et al, 2007), except in the present study the PCR primers were not fluorescently tagged.…”

“…Microsatellites are characterized by short tandem repeats (STRs) of 1 to 6 nucleotide motifs, which widely spread in a random manner throughout the eukaryotic genome. They are commonly used in a wide range of applications, such as the evaluation of genetic diversity and population structure patterns [3,16]. In previous studies, microsatellites have also been used as biomarkers for monitoring heritable traits in rodents [7,33].…”
