Monochamus saltuarius is a vector for pine wilt disease that causes enormous damage to native pine trees in Korea. To develop a biological control method for this pine wilt disease vector, an entomopathogenic fungus was isolated from the cadaver of an adult M. saltuarius supporting fungal conidiation. This fungus was named MsW1 and identified as Beauveria bassiana by microscopic examination, PCR amplification using B. bassiana -specific primers and genetic sequencing of the ITS and EF1-α regions. Virulence tests against M. saltuarius were conducted with conidial suspensions (1 × 10(8) conidia/ml) of B. bassiana MsW1 in laboratory conditions. The median lethal times (LT(50)) of adults and larvae were 7.2 and 7 days, and 100% mortality was observed at 11 and 13 days after inoculation, respectively. This is the first characterization of B. bassiana from M. saltuarius.

Antagonism between entomopathogenic fungi and bacterial symbionts of entomopathogenic nematodes

Mutual effects between the symbiotic bacteria of entomopathogenic nematodes, Photorhabdus luminescens and Xenorhabdus poinarii, and entomopathogenic fungi were investigated in vitro. A dual culture assay on nutrient agar supplemented with bromothymol blue and triphenyltetrazolium chloride (NBTA) medium revealed that P. luminescens is antagonistic to Metarhizium anisopliae, Beauveria bassiana, B. brongniartii and Paecilomyces fumosoroseus by inhibiting their growth and conidial production; the fungal growth was not inhibited by X. poinarii. In a second laboratory experiment, crude extract produced by M. anisopliae was tested for its activity against P. luminescens and X. poinarii. Crude extract from M. anisopliae was antibacterial to P. luminescens and X. poinarii at 1000 lg/ml and inhibited their growth on NBTA, but had no effect at 100 or 10 lg/ml. The influence of the crude extract of M. anisopliae on the dispersal of infective juveniles (IJs) of Heterorhabditis megidis and Steinernema glaseri was assayed on Sabouraud Dextrose Agar (SDA) plates. Results showed that the crude extract of M. anisopliae had no toxic effects even at highest concentration (1000 lg/ml).

The virulence against the two-spotted spider mite, Tetranychus urticae Koch, was evaluated with entomopathogenic fungus Paecilomyces lilacinus (Thom) Samson HY-4 which is isolated from coleopteran insect and registered to Korean and US patents. Virulence tests were conducted with conidial suspensions (1.5 ×10 6 conidia/ml) of P. lilacinus HY-4 against T. urticae adults and positive results were recorded in laboratory conditions. The spraying device was also developed for the efficient and exact evaluation of treatment. The developed spraying device was named as SD-tower sprayer and its efficacy of spraying conidia was evaluated. The accumulated mortality caused by P. lilacinus HY-4 using SD-tower spray was about 73% at 6 days after inoculation. This suggest that the isolate P. lilacinus HY-4 may be considered as promising for a new approach to prevent adult infestations by T. urticae.
