Seed storage protein composition of Hellenic bread wheat cultivars

Abstract: The allelic diversity in seed storage proteins of 25 bread wheat cultivars grown in Hellas was investigated. In total, 15-20 seeds per cultivar were used for the determination of the alleles present at the loci coding for high-molecular-weight glutenin subunits and gliadins. For this purpose, acid polyacrylamide gel electrophoresis for gliadins and SDSelectrophoresis for glutenins were employed. Analysis of the electrophoretic patterns revealed that intravarietal selections obtained from the cultivar ÔNestosÕ,… Show more

“…Central to this discussion is the statement in Article 1 of UPOV, Act 1991, that the distinctness of a variety is determined by the expression of characteristics that describe the genotype or combination of genotypes (UPOV, 1991). In this respect, they are analogous to protein markers currently in use for variety identifi cation in some cereals (Xynias et al, 2006) and potato (Solanum tuberosum L.) (Cooke, 1999), where polymorphisms observed in expressed regions are derived from polymorphisms within the genes. “…The authors of the present study believe that they are a more reliable tool than the biochemical markers. This is because the biochemical markers detected the presence of the translocation in a proportion of plants in cultivar Chios (Xynias et al 2006) something that it is not true. However, if the positive advantages of the translocation are to be exploited more effectively then more sources of the translocation must be utilized in different genetic backgrounds (Dimitrijevic et al 2008).…”

“…Cultivar Chios it was examined in more details, since in a previous research it was found to be consisted of two different genotypes, one carrying the translocation (Xynias et al 2006). For this, 20 individual plants were subjected to PCR.…”
