A biosurfactant-producing bacterium (Staphylococcus sp. strain 1E) was isolated from an Algerian crude oil contaminated soil. Biosurfactant production was tested with different carbon sources using the surface tension measurement and the oil displacement test. Olive oil produced the highest reduction in surface tension (25.9 dynes cm(-1)). Crude oil presented the best substrate for 1E biosurfactant emulsification activity. The biosurfactant produced by strain 1E reduced the growth medium surface tension below 30 dynes cm(-1). This reduction was also obtained in cell-free filtrates. Biosurfactant produced by strain 1E showed stability in a wide range of pH (from 2 to 12), temperature (from 4 to 55 °C) and salinity (from 0 to 300 g l(-1)) variations. The biosurfactant produced by strain 1E belonged to lipopeptide group and also constituted an antibacterial activity againt the pathogenic bacteria such as Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa and Bacillus subtilis. Phenanthrene solubility in water was enhanced by biosurfactant addition. Our results suggest that the 1E biosurfactant has interesting properties for its application in bioremediation of hydrocarbons contaminated sites.

Microorganisms are the important factors in the degradation of the toxic substances in our environment.Petrol and diesel oil is one of the complex mixtures which cannot be easily degraded. The Bacillus cereus was involved in the degradation of oil during which the complex toxic substances were detoxified by the production of biosurfactants. In our study we have identified that the biosurfactant producing Bacillus cereus have a high potential for hydrocarbon degradation. The Bacillus cereus was isolated from hydrocarbon contaminated soil and identified based on morphology and biochemical test according to the Bergey's manual of systematic bacteriology. The maximum hydrocarbon degrading biosurfactant producing Bacillus cereus was obtained by qualitative and quantitative methods. In optimization studies, the best results observed for Bacillus cereus were, Olive oil as the suitable carbon source, Sodium nitrate as the best Nitrogen source and Optimum pH is 7 and Optimum temperature is 37°C. The ability of these isolates to degrade hydrocarbons and survive in the oil contaminated soil is attributed to the development of resistance by mutation on the plasmid. It is also clearly evident that the specific gene was responsible for the production of biosurfactant and the degradation process. According to the results from the present study the Bacillus cereus has high potential for hydrocarbon degradation and can be used especially for Microbial Enhanced Oil Recovery and bioremediation of hydrocarbons in near future.

The bacteria (Brevundumonas diminuta, Pseudomonas fluorescens, Pseudomonas aeroginosa, Pseudomonas citronelbis) which are indigenous bacteria from Babat Toman village South Sumatra. It was isolated and identified by using Bergey's method. In the normal condition (pH 7), these bacteria could produce bio surfactant and effectively as a mixed culture. The growth of a mixed culture of indigenous bacteria was tested at different pH conditions (5-9). The results show that the optimum pH was obtained at pH 7, the population of bacteria was 2.14.10 7 cfu/mL. The shortest regeneration time of these bacteria were (12, 12, 9, 9) hours respectively. The initial total petroleum hydrocarbon concentration of the sample was 18,4 %, it was extracted by using the bio surfactant of mixed culture with 10 % concentration at different pH conditions (5-9), the contact time as long as 168 hours. The optimal oil recovery was obtained at pH 7 as much as 20.97 %. It can be concluded that the bio surfactant effectively used in the normal condition (pH 7) and ineffectively in alkaline condition (pH 9). The GC analyses were conducted to initial crude oil and the residue. The results show that the crude oil consists of 33 components and the residue consists of 14 components. It shows that the bio surfactant of mixed culture bacteria dissolved the hydrocarbon components with temperature fractions 150, 155, 178, 184, 190, and 197 o C.
