Screening Random Peptide Libraries with Subacute Sclerosing PanencephalitisBrain-Derived Recombinant Antibodies Identifies Multiple Epitopes in the C-Terminal Region of the Measles Virus Nucleocapsid Protein

Abstract: Infectious and inflammatory diseases of the CNS are often characterized by a robust B-cell response that manifests as increased intrathecal immunoglobulin G (IgG) synthesis and the presence of oligoclonal bands. We previously used laser capture microdissection and single-cell PCR to analyze the IgG variable regions of plasma cells from the brain of a patient with subacute sclerosing panencephalitis (SSPE). Five of eight human IgG1 recombinant antibodies (rAbs) derived from SSPE brain plasma cell clones recogni… Show more

“…Recombinant antibodies (rAbs) were prepared from single, clonally expanded MS CSF plasma cells, as described. 13 Each rAb was generated from an expanded plasma cell clone defined by identical heavy and light chain variable region sequences in 2 or more plasma cells. 8 Thirty-two rAbs, generated from 4 patients with MS (MS02-19, MS03-1, MS04-2, and MS04-3), and representing 73%-100% of clonally expanded plasma cell populations, were used in immunocytochemistry.…”

“…Currently, traditional clinical diagnosis depending on symptomatic findings often leads to misdiagnosis of measles. Establishment of serological assays for measles is important not only from the standpoint of the prevention of infection, but also confirms the definitive diagnosis [18].…”

“…The measles elimination important in strengthening surveillance and vaccination rates of improvement. Also enhancing epidemiological surveillance and scientific monitoring system based on the characteristics of virus and gene is essential for the eradication of measles (28,29).…”
