Screening and functional prediction of differentially expressed circRNAs in proliferative human aortic smooth muscle cells

Abstract: | INTRODUC TI ONCardiovascular diseases (CVD), including vascular remodelling diseases, are the leading cause of mortality among humans throughout the world. Abnormal proliferation of vascular smooth muscle cells (VSMCs) is a common feature of many vascular remodelling diseases, including atherosclerosis, 1 hypertension 2 and vascular aneurysms. 3 The regulation of VSMC proliferation has been considered a key event due to its major implications for the prevention of pathological vascular conditions. 4 Non-codi… Show more

“…However, searching the ideal biomarker for the accurate diagnosis and effective treatment of PCOS has been a formidable challenge because its pathogenesis has not been fully elucidated. Over the past decade, circRNAs were regarded as novel and vital regulators for the expression of genes as their altered expressions had been strongly associated with a variety of diseases 29‐32 . Accumulating evidences suggest that circRNAs could be used as the excellent indicators of diagnosis and prognosis.…”

“…Using a unique source of 151 aortic SMCs from multiethnic donors, we identified 1,589 circRNAs of which 173 were differentially expressed between quiescent and proliferative SMCs. Although previous studies have performed differential expression of circRNAs between PDGF-BB treated and control vascular SMCs 11,12,23,24 , our study tested differences in circular RNA expression across SMCs cultured in the presence or absence of fetal bovine serum. While we did not treat our RNA with linear RNA degradation (Rnase R) enzyme like these other studies, we included many more samples (150 donors in two conditions).…”

“…Although circRNAs and lncRNAs differ significantly in shape and size, both are expressed in tissue- and spatiotemporal-specific patterns and regulate their target genes by similar mechanisms [ 33 36 ]. Since the ceRNA hypothesis was proposed [ 37 ], both circRNAs and lncRNAs have been reported to function as miRNA sponges that alter the stability or translation of mRNAs [ 21 , 38 ]. Thus, we used microarray and bioinformatics analyses to detect DEcircRNAs and DElncRNAs between the prefrontal cortex tissues of Nrf2 (-/-) and Nrf2 (+/+) mice.…”
