Screening and Characterization of Lactic Acid Bacteria Strains with Anti-inflammatory Activities through in vitro and Caenorhabditis elegans Model Testing

Abstract: The present study was conducted to screen candidate probiotic strains for anti-inflammatory activity. Initially, a nitric oxide (NO) assay was used to test selected candidate probiotic strains for anti-inflammatory activity in cultures of the murine macrophage cell line, RAW 264.7. Then, the in vitro probiotic properties of the strains, including bile tolerance, acid resistance, and growth in skim milk media, were investigated. We also performed an in vitro hydrophobicity test and an intestinal adhesion assay … Show more

“…In the best-studied example of AI-2 inhibition, the brominated furanones isolated from a marine alga, Delisea pulchra, inhibited several multicellular physiologies of bacteria without affecting general growth (Ren et al 2001). LAB are found as commensal microorganisms in the human and animal gastrointestinal (GI) tracts and have a long history of safety and interest in their beneficial properties (Lee et al 2015). “…However, these furanone compounds contain halogens and are toxic; hence, a better approach for food-grade materials is to target QQ activity. LAB are found as commensal microorganisms in the human and animal gastrointestinal (GI) tracts and have a long history of safety and interest in their beneficial properties (Lee et al 2015). However, the impact of LAB on the diversity of the gut microbial community is not well understood.…”

“…After incubation, 10‐fold serial dilutions were made, and then, live cells were counted on MRS agar plates after 24 hr of incubation at 37°C. Tolerance was expressed as a percentage of the number of viable cells after incubation in artificial gastric acid MRS broth relative to that of the normal MRS broth and was calculated using the following equation (Lee, Choi, Lee, Lee, & Park, ):Tolerance(%)=(LogCFU/mlinartificialgastricacidoracidicsolution)/(LogCFU/mlinnormalMRSbroth)×100.…”
