Scintigraphic measurement of regional gastrointestinal transit in the dog

Abstract: Scintigraphic techniques can measure sequentially gastric emptying, small bowel transit, and colonic transit in humans, and comparable methods for experimental studies in animals would be useful. We developed such a method in dogs and examined the effects of prokinetic drugs on regional transit. Two isotopes were given to fasting dogs. Polystyrene pellets labeled with99mTc were mixed in a can of dog food and 111In- labeled pellets were given in a gelatin capsule coated with a pH-sensitive polymer, designed to … Show more

“…Postprandial gastric motility is assessed either by quantifying gastric emptying time and/or gastric antral motility or by detecting the electrical excitement caused by gastric movement. Gastric emptying can be evaluated using scintigraphy [1,14], a 13 Coctanoic acid breath test [8,31], or radiopaque markers [18,27], whereas gastric antral motility is examined using force transducers [13] or abdominal ultrasonography [5,7,9,19].…”

“…A variety of different techniques are available for the assessment of drug-induced changes in GI motility, from the conventional organ bath preparation and the gold standard preclinical in vivo evaluation method, the rodent charcoal meal (Harrison et al 2004), to newer and more innovative methods including the use of wireless motility devices and imaging (Iwanaga et al 1998;Chiba et al 2000Chiba et al , 2002.…”

“…Anterior images were acquired at 0, 15, 30, 45, 60, 90, 120, 180 and 240 min after the meal. The region of interest was drawn around the stomach on the anterior and posterior images for each frame; a semi-auto-analysis system was used to calculate the emptying of the stomach by computer methodology (25).…”
