SARS-CoV-2 Omicron detectionRT-qPCR assay with BA.1 and BA.2/BA.3 differentitation v1

Abstract: Omicron lineage (B.1.1.529) – SARS-CoV-2 variant of concern designated on 26 November 2021 by WHO. This variant has a large number of mutations, some of which are concerning. There is already enough evidence that Omicron lineage connected with increased risk of reinfection with this variant and reduced neutralization by convalescent and vaccinated sera, as compared to other VOCs. Omicron subvariants includes several subvariants with strikingly different genetic characteristics: BA.1, BA.2, BA.3. We developed R… Show more

“…A new RT-qPCR test (using the Δ31-33 amino acid deletion in the N-gene) has recently been developed to specifically detect Omicron and its sub-variants (BA.1, BA.2, and BA.3) with the opportunity to differentiate BA.1 from BA.2/BA.3. [50,51] The Omicron variant presents high mutations in its spike/ RBD regions compared to previous SARS-CoV-2 variants. This could lead to false negative results in the PCR assays that are equipped only to detect the S-gene.…”

“…[10] A multiplex RT-qPCR assay specific to Omicron lineage has been developed recently to detect and differentiate the Omicron subvariants (BA.1, BA.2/BA.3). [11] In addition, the Omicron variant is mainly making severity (oxygen and ventilator requirements) to people of age higher than 70 and the person having comorbidities such as diabetes, blood pressure, cardiac and neurological issues. The symptoms are non-specific as compared to other variants and the symptoms are differently observed in younger and elders.…”
