Sandwich immunoassay for the prostate specific antigen using a micro-fluxgate and magnetic bead labels

Abstract: We describe a micro fluxgate based device with rectangular magnetic core for the determination of prostate specific antigen (PSA) labeled with Dynabeads. A sandwich immunoassay was employed where PSA is captured on a gold film modified with a self-assembled monolayer of antibody. The secondary antibody is labeled with Dynabeads. By applying a DC magnetic fields in the range of 460 to 700 μT, PSA can be detected with detection limit as low as 0.1 ng mL −1 . This micro fluxgate-based assay offers the advantages … Show more

“…Although those methods are reliable, most of them require long measurement procedures, high costs, complex equipment, highly qualified personnel and are not appropriate for massive decentralized assays. Therefore, nowadays it is still a challenge to develop simple, low‐cost and appropriate detection methods for sensitive determination of PSA levels . In this work, a simple and sensitive nanostructured label free immunosensor for PSA quantification is proposed.…”

“…Blood PSA concentration is significantly augmented in prostate cancer patients, because of the disorder of the prostate gland . PSA is used extensively as a prostate cancer biomarker with the aim of screen, diagnose, follow‐up the response to treatment and detection of recurrence after definite therapy . A value of PSA higher than the cut‐off value (2.5–4 ng mL −1 ) indicates high probability of prostate cancer .…”

“…Therefore, some welldeveloped methods for magnetic¯eld detection have been used in biosensing applications, including (i) magnetoresistive (MR) methods such as anisotropic magnetoresistance (AMR), [46][47][48][49][50][51][52] giant magnetoresistance (GMR), [53][54][55][56][57] tunneling magnetoresistance (TMR), 58,59 giant magnetoimpedance (GMI) [60][61][62][63] and (ii) nonMR methods such as those involving the use of a°ux gate sensor, Hall sensor and superconducting quantum interference device (SQUID). [64][65][66][67][68][69] Several magnetic¯eld sensors have been used in biodiagnostics, but the drawbacks of large size, low sensitivity, or high power consumption limited their use in practical applications. 70 The use of magnetic sensors based on MR e®ects is a promising alternative to overcoming these concerns related to nonMR sensors.…”

“…For example, the micro°uxgate sensor is used to trap and detect speci¯c antigens. 69 When a uniform magnetic¯eld is applied in the out-of-plane direction of a°at rectangular semiconductor and in which electric current°ows along the long bar, voltage is then generated across the width of the°at plate, as shown in Fig. 6(a).…”
