S-glutathionylation, friend or foe in cardiovascular health and disease

Abstract: Glutathione is a low molecular weight thiol that is present at high levels in the cell. The high levels of glutathione in the cell make it one of the most abundant antioxidants contributing to cellular redox homeostasis. As a general rule, throughout cardiovascular disease and progression there is an imbalance in redox homeostasis characterized by reactive oxygen species overproduction and glutathione underproduction. As research into these imbalances continues, glutathione concentrations are increasingly bein… Show more

“…These extreme conditions are rare in vivo. Therefore, for most proteins, the spontaneous formation of Pr-SSG—as a result of the exchange of Pr-SH and GSSG—is not common and, as a rule, takes place under pathological conditions [ 30 ]. In addition, a variant of thiol-disulfide exchange between Pr-SH and a protein which is already S-glutathionylated (Pr′-SSG) is possible: …”

“…The metabolic cofactor glutathione (GSH) is considered the most abundant endogenous antioxidant (discovered more than 10 decades ago) that plays an important role in detoxification of reactions in CCs [ 72 ]. This is a tripeptide that is biosynthesized in two key steps.…”

“…Glrx reverses S-glutathionylation of GAPDH and plays a role in the key step of apoptosis signal transduction, thus participating in regulating apoptosis [ 40 ]. Moreover, eNOS is regulated by direct S-glutathionylation in the endothelial cells [ 41 ]. NO reacts with superoxide to form peroxynitrite, which activates S-glutathionylation in Cys674 to regulate SERCA2 activity and vascular smooth muscle function [ 42 ].…”
