Rutin promotes the formation and osteogenic differentiation of human periodontal ligament stem cell sheets invitro

Abstract: Cell sheet technology is a novel tissue engineering technology that has been rapidly developed in recent years. As a novel technology, cell sheet technology is expected to become one of the preferred methods for cell transplantation. The present study investigated the biological effects of rutin on the formation of periodontal ligament stem cell (PDLSC) sheets and their resultant osteogenic properties. The results of Cell Counting Kit-8 (CCK-8) assay demonstrated that a concentration of 1×10−6 mol/l rutin prom… Show more

“…All participants provided signed informed consent in accordance with the Helsinki Declaration. The method of hPDLSC isolation from teeth was similar to a previously reported one ( 31 ). Briefly, the extracted teeth were placed in a 15 ml centrifuge tube with α-MEM (Biological Industries) amended with 5% antibiotics (100 U/ml penicillin, 10 mg/ml streptomycin) on ice.…”

“…periodontal curettage and root planning) and guided tissue regeneration [ 12 ]. Periodontal curettage can control the progression of periodontitis and prevent the further loss of periodontal support tissue [ 13 ]. Root planing is widely used procedures for treatment of periodontitis, due to their low cost and effectiveness in reducing the clinical signs of inflammation and the levels of pathogens [ 14 , 15 ].…”

“…Among these, TNF-α is an important member of the TNF family, showing its effect primarily through the NF-κB or MAPK signaling pathways. It is also the primary inflammatory factor regulating tissue destruction in periodontitis, and is involved in several different inflammatory reactions ( 27 ).…”
