Rust and Downy Mildew Resistance in Pearl Millet (Pennisetum glaucum) Mediated by Heterologous Expression of the afp Gene from Aspergillus giganteus

Abstract: The cDNA encoding the antifungal protein AFP from the mould Aspergillus giganteus was introduced into two pearl millet (Pennisetum glaucum) genotypes by particle bombardment. Stable integration and expression of the afp gene was confirmed in two independent transgenic T0 plants and their progeny using Southern blot and RT-PCR analysis. In vitro infection of detached leaves and in vivo inoculation of whole plants with the basidomycete Puccinia substriata, the causal agent of rust disease, and the oomycete Scler… Show more

“…tritici infections [17]. Reduced symptoms of plant diseases were observed in transgenic rice after M. grisea infection [18,19], and in transgenic pearl millet infected with the downy mildew Sclerospora graminicola or the rust Puccinia substriata [20]. AFP also prevented the infection of Lycopersicon esculentum roots with Fusarium oxysporum f. sp.…”

“…Transgenic rice constitutively expressing AFP protein showed inheritance of the transgene without any effect on plant morphology, growth and development (Moreno et al, 2005). Recently, rust and downy mildew resistance in pearl millet (Pennisetum glaucum) was achieved by heterologous expression of the afp gene from Aspergillus giganteus through particle bombardment of immature zygotic embryos (Girgji et al, 2006). However, examples of antifungal peptides from microorganism origin expressed in plants are still scarces and one of the unexplored niches.…”

“…The crop is grown worldwide mainly for food and forage (Girgi et al, 2006). It performs well under stressful conditions of drought and acidic soils (FAO, 2004); though it also does competitively well in favorable environments (Bhatnagar et al, 1998;Christinck, 2002).…”
