RSM-optimized IRAE sample pretreatment and HPLC simultaneous determination of Tryptanthrin, indigo, and indirubin from Chinese herbal medicineRadix Isatidis

Abstract: Summary.Radix Isatidis has widely useful activities including anti-virus, anti-bacterial. Tryptanthrin, indigo, and indirubin are active ingredients in R. Isatidis. Response surface methodology (RSM)-optimized infrared-assisted extraction (IRAE) was developed and combined with HPLC for simultaneous determination of tryptanthrin, indigo, and indirubin from R. Isatidis. IRAE were investigated through extraction yields of the three components and optimized by RSM. The optimum conditions were as follows: infrared … Show more

“…About 6.0 g of root bark, root core, stem bark, stem core, near-leaf stem or leaf of T. asiatica was soaked separately in methanol for 24 h at a solid-liquid ratio of 1:50 (g:mL), extracted three times by reflux method for 2 h each time in a 65 8C-constant temperature water bath [9,11,21]. Six extracts from different parts of T. asiatica were finally prepared.…”

“…9,10) The most common and traditional procedures for TCMs are solvent extraction, Soxhlet extraction, heat-reflux extraction, and ultrasound-assisted extraction, which are most widely used currently, but these methods involve large solvent consumption or long extraction time. [11][12][13][14][15][16] New techniques like microwave-assisted extraction and supercritical fluid extraction are alternative method which can also be employed. [17][18][19][20][21] However, both of these two pretreatment techniques need high-cost specific equipment.…”

“… 8 Tryptanthrin as a highly selective cyclooxygenase-2 inhibitor showed inhibitory activity on prostaglandin and leukotriene synthesis, and reduced leukotriene B4 levels with a potency comparable to that of the clinically used 5-lipoxygenase inhibitor 10 , 11 and so on. Because of its very low content in plants, less than one ten-thousandth (mass/crude drug mass), 2 , 15 and the fact that it is difficult, laborious, time-consuming, and inefficient in separation and purification of such complex natural samples, targeted synthetic methods were preferred to obtain this potential drug through microbial fermentation, 16 biomimetic synthesis 17 and chemical synthetic methods. 18 , 19 , 20 The pharmaceutical analysis method, an important tool for tryptanthrin drug development and clinical drug evaluation, can timely reflect the problems arising in the drug production process.…”
