RpoE Facilitates Stress-Resistance, Invasion, and Pathogenicity of Escherichia coli K1

Abstract: Escherichia coli K1 is the most common Gram-negative bacterium that causes neonatal meningitis; thus, a better understanding of its pathogenic molecular mechanisms is critical. However, the mechanisms by which E. coli K1 senses the signals of the host and expresses toxins for survival are poorly understood. As an extracytoplasmic function sigma factor, RpoE controls a wide range of pathogenesis-associated pathways in response to environmental stress. We found that the ΔrpoE mutant strain reduced the binding an… Show more

“…Pelagibacter’ HTCC7211, TMED112 CCGs have lost the two‐component system (EnvZ‐OmpR; ChvG‐ChvI; NtrY‐NtrX and RegB‐RegA) related to nitrogen regulation, acidity sensing, osmotic stress, and redox response. Regarding sigma factors, while strain HTCC7211 had two ( rpo D and rpo H), TMED112 CCGs also encode rpo E, involved in the response to several environmental stresses (Fan et al, 2022).…”

“…The phosphate-sensing PhoR-PhoB system has also been associated with E. coli virulence and is required for expression of a gene that regulates type I pili synthesis in CFT073 ( 56 ) ( 57 , 58 ). RpoE has been implicated in virulence of Salmonella and E. coli , including UPEC strains ( 59 62 ). RpoE is stress-induced sigma subunit for RNA polymerase that responds to high temperature and osmolarity, misfolded proteins in the outer membrane, periplasm, and inner membrane, metal ion exposure, entry into stationary phase, and elevated (p)ppGpp ( 40 ).…”

“…RpoE directly regulates expression of some virulence genes such as ompA, cnf1, fimB, ibeA , and the capsule genes kpsM , and kpsF . Also, RpoE indirectly modulates some virulence genes such as fimA, aslA , and traJ (Fan et al 2022 ). This example shows the broad impact of SR on regulation of virulence during bacteremia.…”
