A simple, rapid, and precise method is developed for the quantitative simultaneous estimation of telmisartan and hydrochlorothiazide in combined pharmaceutical dosage form. A chromatographic separation of the two drugs was achieved with an ACE 5 C 18 (250 x 4.6 mm) analytical column using buffer-acetonitrile (55:45 v/v). The buffer used in mobile phase contains 0.1M sodium perchlorate monohydrate in double distilled water pH adjusted 3.0 with trifluoroacetic acid. The instrumental settings are flow rate of 1.5 ml min -1 , column temperature at 30 o C, and detector wavelength of 215 nm using a photodiode array detector. The resolution between hydrochlorothiazide and telmisartan founds to be more than 5. Theoretical plates for hydrochlorothiazide and telmisartan were 13022 and 6629 respectively. Tailing factor for hydrochlorothiazide and telmisartan was 0.94 and 0.98 respectively. Telmisartan, hydrochlorothiazide and their combination drug products stressed samples were analysed by the proposed method. The described method shows excellent linearity over a range of 70 to 130% of target analyte concentration. The correlation coefficient for telmisartan and hydrochlorothiazide are 0.9999. The relative standard deviation for six measurements in two sets of each drug in tablets is always less than 2%. The proposed method was found to be suitable and accurate for quantitative determination and stability study of telmisartan and hydrochlorothiazide in pharmaceutical preparations.

A simple, rapid, precise, sensitive and reproducible reverse phase high performance liquid chromatographic (RP-HPLC) method for determination of Telmisartan (TELM) in tablet dosage form was developed and validated. Chromatographic separation was achieved on a 250 × 4.6 mm, 5µ, Waters symmetry column in gradient mode, with mobile phase consisting of a mixture of solution (10 mM potassium dihydrogen phosphate, pH 3.5 ± 0.01): acetonitrile (64:40) was used. The quantitation performed at flow rate of 1.0 mL/min at 230 nm and run time was 12 min. The analytical method was validated as per ICH guideline for linearity, accuracy, precision, specificity, limit of detection, limit of quantification, robustness and stability and method can be extended to the analysis of TELM in tablet formulations. The relative standard deviation values for precision was less than 2%, and % recovery was greater than 98% for TELM. The drug undergoes oxidative degradation, thermal degradation and in alkali medium.

Drugs are traditionally been identified on basis of chromatographic-spectroscopic hyphenated techniques in instrumental analysis. Gas chromatography (GC) and Liquid chromatography (LC) hyphenated with mass spectroscopy (MS) i.e. GC-MS and LC-MS give reliable and confirmatory results in drugs identification. In the present work the novel hyphenated technique High Performance Thin Layer Chromatography-Mass Spectroscopy (HPTLC-MS) has been used. This technique provides efficient, quick and simple method for identification and separation of Narcotic drugs and psychotropic substances. The drugs under study are Papaverine, Methadone, Cocaine, Ketamine, Caffeine, Codeine, Diazepam, Thebaine, Heroin, Methamphetamine, Carbamazepine, Morphine, Narcotine and Ephedrine. The present study comprising of sixteen drugs has been carried out on CAMAG HPTLC instrument with automatic sampling. Thin layer chromatography (TLC) plates were developed in various solvent systems, scanned under TLC scanner and the results in terms of Retention Factor (R f value) and UV spectrum (λmax) are presented in the manuscript. Using hyphenated technique of HPTLC-MS (MS 2020 SHIMADZU) spots of these drugs from TLC plate was lifted with CAMAG TLC-MS interface and confirmed by the mass spectrum of the individual drugs by their m/z values thus delivering fast and accurate confirmatory result on the TLC plate.
