Rotenoid Derivatives from Derris Trifoliata with Nitric Oxide Production Inhibitory Activity

Abstract: Study of the chemical constituents of the stems of Derris trifoliata Lour. (Leguminosae) collected in Singapore led to the isolation and identification of three known and two new rotenoid derivatives. The new derivatives, named derrisfolin A (1) and B (2), inhibited nitric oxide production in murine macrophage-like RAW 264.7 cells stimulated with interferon-γ and lipopolysaccharide.

“…We previously reported that derrisfolin A (Fig. S1 ), our isolated novel natural compound, inhibited the excess production of the proinflammatory mediator NO in RAW264.7 macrophages stimulated with LPS and interferon‐γ [ 38 ]. We used microarray analysis to determine changes in gene expression in derrisfolin A‐treated MIN6 cells and RAW264.7 macrophages, and we found that RGN gene expression was dramatically upregulated among 12 differentially expressed genes, with > 10‐fold changes in the two‐type cells (unpublished data).…”

“…The RAW264.7 mouse macrophage cell line was maintained in RPMI 1640 medium containing 10% heat-inactivated FBS, 50 UÁmL −1 penicillin, and 50 μgÁmL −1 streptomycin under a humidified atmosphere of 5% CO 2 at 37 °C. The derrisfolin A used in this study was isolated and purified as previously described [38]. Derrisfolin A purity was assessed by thin-layer chromatography and 13 C nuclear magnetic resonance.…”

“…[5][6][7][8] These types of compounds have been shown to have potential cytotoxic and anti-inflammatory activities. 9,10 The roots of D elliptica contain rotenoids, such as rotenone, deguelin, and tephrosin, which are toxic ingredients responsibility for the plant's insecticidal property. 5,7 Other phenolic compounds, including flavone, isoflavone, pterocarpan, and cumaronochromone, have been revealed from the aerial parts of this plant.…”
