Jatropha platyphylla is considered as a potential source of edible protein, oil, and phenolic compounds with anti-inflammatory activity. The use of stem cutting in vegetative propagation and grafting is as indispensable tools for mass multiplication of superior genotypes and helps in improve planting yield and quality. The study was aimed to investigate the effect of different diameters (10 -15, 16 -25 and 26 -35 mm) and different hormone concentrations of indo-butyric acid (0, 1.5, 3, 6 and 10 g/L), in the rooting of Jatropha platyphylla and to obtain axillary buds to performed grafts. Rooting efficiency was 80% in greenhouse conditions. Hormone concentration and diameter significantly affected the rooting and shooting ability of Jatropha platyphylla stem cuttings. Stem cuttings of 26 -35 mm have the greatest number, length and dry root weight. 76% survival of the grafted plants was obtained. This demonstrates the necessity to improve the conventional propagation methods with appropriate scientific inputs for more economical and time efficient techniques for standard propagation protocols.

Background and Objective: Macrotyloma uniflorum is nutritious and a good source of bioactive substances with high biological processes. The high frequency and efficient in vitro regeneration protocol have developed first time by utilizing embryo and ovary explants in Horsegram, Macrotyloma uniflorum. Materials and Methods: This research was carried out from horse gram paiyur-2 variety seeds derived embryo and ovary explants used by MS medium supplemented with cytokinins and auxins. Results: The phytohormone 0.4 mg LG 1 zeatin induced a maximum of 12 and 9 shoots from an embryo and ovary explants, respectively. The maximum frequency of rooting with about 9 roots was procured from the shoots cultured from embryonic explants using 0.2 mg LG 1 NAA and 7 roots were propagated from shoots derived from ovary explants. Overall studies observed that the embryonic explants show a high percent frequency of shoot and root induction when compared to ovary culture and further plantlets rose using embryonic explants with shoot growth period than the ovary explants. Conclusion: In vitro legume regeneration utilizing embryo and ovary is efficient, dependable and has fewer problems. Embryo explants were used to induce high-frequency plant regeneration, indicating that the ovaries are useful in genetic transformation.

Pedilanthus tithymaloids is a popular temperate indoor succulent ornamental plant and is highly marketable. However, the appropriate propagation techniques should be adopted to solve the rooting issues of cuttings. Therefore, the present research was designed to specify the optimal indole − 3−butyric acid (IBA) concentration and rooting medium type for obtaining good rooting and healthy plants. Three IBA levels (0, 1.0, 2.0, and 3.0 g L− 1) and five growing mediums (sand, sand: clay, sand: peat moss, sand: perlite, and sand: vermiculite as 1:1, v/v) were used. The experiment was performed in a randomized complete block design in three replications. The obtained data illustrated that roots number per cutting and rooting percentage reached the maximum values by treating P. tithymaloids cuttings with 3.0 g L− 1 IBA x sand: perlite or sand: vermiculite medium. Cultivating the treated-IBA 3.0 g L− 1 cuttings of P. tithymaloids in sand: perlite showed the most enhancements in plant height, branches number and leaves number, leaf area and herb dry weight. However, the heaviest root dry weight was noticed with IBA 3.0 g L− 1 x sand: peat moss medium. Except sand medium, all other tested media in combination with IBA at 1.0, 2.0–3.0 g L− 1 showed remarkable stimulation effect on chlorophyll content. Sand: vermiculite mixture x IBA at 2.0 g L− 1 was the effective practice for improving carbohydrates percentage. By studying the anatomical changes of the adventitious root of P. tithymaloids plant, it was found that there was an increase in the root diameter, phloem and xylem tissues and xylem vessels diameter due to growing in sand: perlite x 3.0 g L− 1 IBA. In conclusion, dipping the cuttings bases of P. tithymaloids in 3.0 g L− 1 IBA and cultivation in sand: perlite considered the most effective combination practice for better rooting ability and growth and could be advisable for commercial production.
